Adult flies expressing Hvir\HR13Scer\UAS.cKa under the control of two copies of Scer\GAL4Or67d-GAL4-1 cannot detect the moth Heliothis virescens sex pheromone (Z)-11-hexadecenal if they are also transheterozygous for Snmp11/Snmp12.

The failure of adult flies transheterozygous for Snmp11/Snmp12 to detect the moth Heliothis virescens sex pheromone (Z)-11-hexadecenal when they are also ectopically expressing Hvir\HR13Scer\UAS.cKa under the control of two copies of Scer\GAL4Or67d-GAL4-1 can be partially rescued by co-expression of Mmus\Cd36WT.Scer\UAS : the Or67d neurons now display significant activity (measured by extracellular recordings in individual sensilla) upon (Z)-11-hexadecenal presentation.

The failure of adult flies transheterozygous for Snmp11/Snmp12 to detect the moth Heliothis virescens sex pheromone (Z)-11-hexadecenal when they are also ectopically expressing Hvir\HR13Scer\UAS.cKa under the control of two copies of Scer\GAL4Or67d-GAL4-1 can be restored by co-expression of Snmp1WT.Scer\UAS or Snmp1A401Y.Scer\UAS or Snmp1V353Y.Scer\UAS or (to a lesser extent) Snmp1T274Y.L439Y.Scer\UAS : The Or67d neurons display significant activity (measured by extracellular recordings in individual sensilla) upon (Z)-11-hexadecenal presentation when co-expressing any of the aforementioned Snmp1 transgenes.

The response to the H.virescens (Z)-11-hexadecenal pheromone that is seen in olfactory sensory neurons expressing Hvir\HR13Scer\UAS.cKa under the control of Scer\GAL4Or67d.PF in a wild-type background is almost completely abolished by the addition of Snmp1/Snmp2.

Flies in which the endogenous Or67d receptors in the olfactory receptor neurons have been replaced by the H.virescens Hvir\HR13 receptor (by using flies expressing Hvir\HR13Scer\UAS.cKa under the control of Scer\GAL4Or67d-GAL4-1 in a Or67dGAL4-1 background) show responses to the H.virescens (Z)-11-hexadecenal pheromone that are similar to the responses shown by wild-type flies in response to the D.melanogaster cVA ([Z]-11-octadecenyl acetate) pheromone (which normally activates Or67d); in these mutant flies the T1 trichoid sensilla show a rapid firing response to (Z)-11-hexadecenal and males show suppression of courtship of virgin wild-type females that have been treated with (Z)-11-hexadecenal.
