Snmp11/Snmp12 transheterozygote adult males display impaired detection of the male pheromone cis-vaccenyl acetate, cVA, compared to controls: the Or67d olfactory sensory neurons show almost complete lack of response to cVA presentation (measured by measured by extracellular recordings in individual sensilla).

Mutants have no gross morphological or locomotor defects.

The axonal projections to the antennal lobe of Snmp1/Snmp2 Or67d-expressing olfactory sensory neurons are wild type.

Snmp1/Snmp2 Or67d-expressing olfactory sensory neurons show an increase in spontaneous activity compared to control neurons. The mutant neurons do not show an electrophysiological response after stimulation with cis-vaccenyl acetate (cVA), in contrast to wild-type controls.

The impaired male pheromone (cis-vaccenyl acetate, cVA) detection characteristic for Snmp11/Snmp12 adult males is partially rescued by expression of either ninaD::Snmp1Scer\UAS.T:Avic\GFP-EGFP or ninaDScer\UAS.T:Avic\GFP-EGFP under the control of Scer\GAL4Or67d.PF : the lack of Or67d neuronal activity (measured by extracellular recordings in individual sensilla) is significantly restored. In the case of ninaDScer\UAS.T:Avic\GFP-EGFP the rescue effect is weak and only observed when the cVA is delivered in close range (as opposed to in an air-stream - in that assay no rescue is observed at all).

The impaired male pheromone (cis-vaccenyl acetate, cVA) detection characteristic for Snmp11/Snmp12 adult males is partially rescued by expression of under the control of Scer\GAL4Or67d.PF : the lack of Or67d neuronal activity (measured by extracellular recordings in individual sensilla) is significantly, albeit not fully, restored.

lush1 Snmp1/lush1 Snmp2 double mutant Or67d-expressing olfactory sensory neurons show an increase in spontaneous activity compared to control neurons, as is seen in Snmp1/Snmp2 single mutant neurons.

Snmp1 Or83b2/Snmp2 Or83b1 double mutant Or67d-expressing olfactory sensory neurons show show severely decreased spontaneous activity compared to control neurons, as is seen in Or83b1/Or83b2 single mutant neurons.

Olfactory sensory neurons expressing Or22aScer\UAS.cLb under the control of Scer\GAL4Or67d.PF still show the ectopic responses to a panel of known Or22a ligands (ethyl butyrate, methyl butyrate, pentyl acetate, methyl hexanoate and methyl octanoate) in a Snmp1/Snmp2 background, but the endogenous response of these neurons to cis-vaccenyl acetate is lost in the Snmp1/Snmp2 background.

The impaired male pheromone (cis-vaccenyl acetate, cVA) detection characteristic for Snmp11/Snmp12 adult males is partially rescued by expression of any of the following: Apol\Snmp1Scer\UAS.T:Avic\GFP-EGFP, Amel\Snmp1Scer\UAS.T:Avic\GFP-EGFP, or Mmus\Cd36Scer\UAS.T:Avic\GFP-EGFP under the control of Scer\GAL4Or67d.PF : the lack of Or67d neuronal activity (measured by extracellular recordings in individual sensilla) is significantly, albeit not fully, restored.

Adult flies expressing Hvir\HR13Scer\UAS.cKa under the control of two copies of Scer\GAL4Or67d-GAL4-1 cannot detect the moth Heliothis virescens sex pheromone (Z)-11-hexadecenal if they are also transheterozygous for Snmp11/Snmp12.

The failure of adult flies transheterozygous for Snmp11/Snmp12 to detect the moth Heliothis virescens sex pheromone (Z)-11-hexadecenal when they are also ectopically expressing Hvir\HR13Scer\UAS.cKa under the control of two copies of Scer\GAL4Or67d-GAL4-1 can be partially rescued by co-expression of Mmus\Cd36WT.Scer\UAS : the Or67d neurons now display significant activity (measured by extracellular recordings in individual sensilla) upon (Z)-11-hexadecenal presentation.

The failure of adult flies transheterozygous for Snmp11/Snmp12 to detect the moth Heliothis virescens sex pheromone (Z)-11-hexadecenal when they are also ectopically expressing Hvir\HR13Scer\UAS.cKa under the control of two copies of Scer\GAL4Or67d-GAL4-1 can be restored by co-expression of Snmp1WT.Scer\UAS or Snmp1A401Y.Scer\UAS or Snmp1V353Y.Scer\UAS or (to a lesser extent) Snmp1T274Y.L439Y.Scer\UAS : The Or67d neurons display significant activity (measured by extracellular recordings in individual sensilla) upon (Z)-11-hexadecenal presentation when co-expressing any of the aforementioned Snmp1 transgenes.

The response to the H.virescens (Z)-11-hexadecenal pheromone that is seen in olfactory sensory neurons expressing Hvir\HR13Scer\UAS.cKa under the control of Scer\GAL4Or67d.PF in a wild-type background is almost completely abolished by the addition of Snmp1/Snmp2.
