Expression of one copy of CkIIαdsRNA.Scer\UAS under the control of Scer\GAL4sca-537.4 does not perturb the eye and leads to subtle, yet reproducible, defects in interommatidial bristle patterning. Expression of two copies of CkIIαdsRNA.Scer\UAS, driven by two copies of Scer\GAL4sca-537.4, does not affect ommatidia but leads to more severe bristle defects, such as ectopic, split and missing bristles. Expression of this transgene in a CkIIαTik/+ background does not lead to more extreme eye phenotypes.
Expression of CkIIαdsRNA.Scer\UAS under the control of Scer\GAL4ey.3.5.Exel results in rough eyes with defective interommatidial bristle patterning in the posterior part of the eye when flies are raised at 25oC, but results in wild-type eyes when flies are raised at 18oC. Temperature shifts from 18oC to 25oC at different stages of development show that expression of CkIIαdsRNA.Scer\UAS in the period that overlaps with the onset of R8 patterning (192 to 216 hours after egg laying) is sufficient to produce the rough eye phenotype, while expression before and after this period is not.
Expression of CkIIαdsRNA.Scer\UAS under the control of Scer\GAL4ey.3.5.Exel in a CkIIαTik/+ background results in a more severe eye phenotype than expression in a wild-type background. Scer\GAL4ey.3.5.Exel>CkIIαdsRNA.Scer\UAS, CkIIαTik/+ eyes are rough in all sections, slightly reduced in size, display fused ommatidia, and have an interommatidial bristle patterning that is more defective.
Eye discs that express CkIIαdsRNA.Scer\UAS under the control of Scer\GAL4ey.3.5.Exel display supernumerary R8 photoreceptor cells at the posterior margin of the morphogenetic furrow.
Expression of CkIIαdsRNA.Scer\UAS under the control of Scer\GAL4E(spl).Exel or Scer\GAL4GMR.PF does not cause ommatidial or interommatidial bristle defects.
Expression of CkIIαdsRNA.Scer\UAS under the control of Scer\GAL4sca-537.4 results in the formation of, typically, one extra bristle on the scutellum compared to wild-type flies. Expression with Scer\GAL4455.2 also leads to the formation of extra scutellar bristles. Expression with Scer\GAL4E(spl).Exel leads to the most severe bristle defect, with extra bristles present on both the scutellum and the thorax.
Expression of one copy of CkIIαdsRNA.Scer\UAS under the control of Scer\GAL4sca-537.4 in a CkIIαTik/+ background results in split bristles and branching of the shaft cell. While this phenotype is not seen in a wild-type or CkIIαTikR/+ background, expression of two copies of CkIIαdsRNA.Scer\UAS under the control of one copy of Scer\GAL4<up>sca-537.4 does result in 'branched' bristles in a wild-type background. Expression of two copies of CkIIαdsRNA.Scer\UAS, under the control of two copies of Scer\GAL4sca-537.4, results in ectopic, split and missing macrochaetes and occasionally in split microchaetes.
