A 2.5kb fragment from upstream of the TrpA1 start codon is fused to sequence encoding a GAL4 driver. This is followed by a 1.7kb fragment consisting of TrpA1 sequences from exon 1 to the fourth intron.

ScerGAL4TrpA1.SH is expressed in the 4 AC thermosensory neurons, in 10 other cells in the brain and subesophageal ganglion and in 8 cells in the thoracico-abdominal ganglion. Co-expression of ScerGAL4TrpA1.SH with pre- and postsynaptic markers labels presynaptic terminals in the dorsal protocerebrum and postsynapses in the cell body of the AC neurons, at the subesophageal ganglion and at the dorsolateral protocerebrum.

Inhibition (using Ctet\tetXTNT-LC.UAS) of Scer\GAL4TrpA1.SH cells in adults significantly reduces aversive temperature, but not shock, olfactory memory, compared to controls.

Inhibition (using Hsap\KCNJ2UAS.EGFP) of Scer\GAL4TrpA1.SH cells, in combination with a Gal80[ts] transgene to restrict expression to the adult stage, significantly reduces aversive temperature olfactory memory, compared to controls.
