Expression of BaceScer\UAS.cCSa under the control of Scer\GAL4VT064246 (using tub-Gal80[ts] to limit the expression to adulthood) does not impair either middle- or long-term memory tested in an aversive olfactory memory assay 2hr and 24hr post-training, respectively.

Adult specific expression of BaceScer\UAS.cCSa driven by Scer\GAL4Mef2.247.Switch (only activated when adult flies are fed RU486) or Scer\GAL4c739 (using the TARGET system) does not significantly affect short term memory (2hrs after training) in an olfactory aversive conditioning paradigm (performance index is similar to controls).

28 day old flies expressing BaceScer\UAS.cCSa under the control of Scer\GAL4GMR.PU show vacuole formation in the lamina and the retina. The vacuoles in the lamina are not restricted to the lamina cortex.

No increase in vacuolisation is seen in the brains of 5 days old flies expressing BaceScer\UAS.cCSa under the control of Scer\GAL4Appl.G1a.

20 day old flies expressing BaceScer\UAS.cCSa under the control of Scer\GAL4elav-C155 have a significantly lower performance index in a fast phototaxis assay than young flies or age-matched control flies.

Flies simultaneously expressing ApplScer\UAS.cTa and BaceScer\UAS.cCSa under the control of Scer\GAL4VT064246 display deficit in both middle- and long-term memory and this can be rescued by co-expression of Nep1EY21255.

The impaired middle- or long-term term memory tested in an aversive olfactory memory assay 2hr or 24hr post-training, respectively, characteristic for adult flies expressing Nep1GD11797 under the control of Scer\GAL4VT064246 (combined with tub-Gal80[ts] to limit expression to adulthood) are exacerbated further by co-expression of BaceScer\UAS.cCSa.

Adult specific expression of BaceScer\UAS.cCSa driven by Scer\GAL4Mef2.247.Switch (activated in adult flies fed RU486) significantly enhances short term memory defects in an olfactory aversive conditioning paradigm compared to Appld/+ flies or to controls not fed RU486; there are no significant effects on learning, olfactory acuity or shock reactivity.

Adult specific expression of BaceScer\UAS.cCSa driven by Scer\GAL4c739 (using the TARGET system) significantly enhances short term memory defects in an olfactory aversive conditioning paradigm compared to Appld/+ flies or to genetic or temperature controls; there are no significant effects on learning, olfactory acuity or shock reactivity.

Expression of BaceScer\UAS.cCSa under the control of Scer\GAL4Appl.G1a enhances the increase in vacuolisation seen in the brains of SNF4Aγloe mutant third instar larvae.

Co-expression of BaceScer\UAS.cCSa results in a marked increase in the amyloid deposits seen in flies expressing ApplScer\UAS.cCSa under the control of Scer\GAL4GMR.PF.

Co-expression of BaceScer\UAS.cCSa and ApplScer\UAS.cCSa under the control of Scer\GAL4Appl.G1a results in the formation of numerous toluidine blue-positive deposits in the brain. These flies also have spongiform lesions scattered throughout the brain.

20 day old flies co-expressing BaceScer\UAS.cCSa and ApplScer\UAS.cCSa under the control of Scer\GAL4Appl.G1a have fibrillar structures and dense bodies in the axons.

Comments

Co-expression of BaceScer\UAS.cCSa partially rescues the lethality caused by expression of BaceGD5366 under the control of Scer\GAL4αTub84B.PL.

Co-expression of BaceScer\UAS.cCSa reduces the vacuolisation in the lamina neuropil and cortex of 28 day old flies expressing BaceGD5366 under the control of Scer\GAL4GMR.PU to the levels seen in controls.

Expression of BaceScer\UAS.cCSa under the control of Scer\GAL4elav.PU significantly reduces the vacuolisation of the lamina neuropil and cortex which is seen in 28 day old Bace2045/Df(2L)Exel7038 flies.

Expression of BaceScer\UAS.cCSa under the control of Scer\GAL4Bace.5.4 completely rescues the vacuolisation of the lamina neuropil and cortex which is seen in 28 day old Bace2045/Df(2L)Exel7038 flies.
