Adult flies expressing ManfScer\UAS.cPa under the control of Scer\GAL4Hml.Δ show improved eye tissue regeneration capacity upon UV radiation induced damage (inflicted during pupal stage) compared to wild-type.

The number of lamellocytes in hemocyte smears from third instar larvae expressing ManfScer\UAS.cPa under the control of Scer\GAL4Hml.Δ is significantly increased compared to wild-type.

Expression of ManfUAS.cPa under the control of Scer\GAL4elav.PLu, Scer\GAL4repo or Scer\GAL4ple.PF does not affect the number of dopaminergic neurons in any studied cluster (PAL, PPL1, PPL2ab, PMM1/2 or PPM3) of the adult brain; these animals also develop normally and live for at least 30 days.

The increased number of lamellocytes in hemocyte smears from third instar larvae as well as the adult eye tissue repair defects characteristic for flies expressing ManfScer\UAS.cPa under the control of Scer\GAL4Hml.Δ is suppressed by co-expression of KdelRGD1228.

The increased level of eye tissue degeneration upon UV irradiation (administered during pupal stage) characteristic for adult flies expressing PvrKK101575 under the control of Scer\GAL4Hml.Δ can be rescued by co-expression of ManfScer\UAS.cPa.

Expression of ManfScer\UAS.cPa under the control of Scer\GAL4Hml.Δ reduces the severity of the retinal degeneration phenotype characteristic for adult flies expressing either grimGMR.PC or ninaEG69D.GMR.

Comments

The increased severity of eye degeneration after exposure of one eye to UV irradiation during pupal stage characteristic for ManfΔ96/+ or ManfΔ112/+ adults can be rescued by expression of ManfScer\UAS.cPa under the control of Scer\GAL4Hml.Δ.

ManfScer\UAS.cPa expression driven by Scer\GAL4da.G32 rescues the early larval lethality of ManfΔ96 mutants and normalizes ManfΔ96 larval dopamine to wild type levels.

ManfScer\UAS.cPa expression driven by Scer\GAL469B rescues the early larval lethality of ManfΔ96 mutants.

Expression of ManfScer\UAS.cPa under the control of one of Scer\GAL4da.G32, Scer\GAL4tub or Scer\GAL469B rescues the larval lethality of ManfΔ96 animals (assayed by survival into the pupal stage).

Expression of ManfScer\UAS.cPa under the control of Scer\GAL469B rescues the embryonic lethality of ManfΔ96 embryos that lack both maternal and zygotic Manf[+] function.
