Expression of Hsap\PINK1Scer\UAS.cYa results in a complex interaction with the Hsap\LRRK2Scer\UAS.cVa and Hsap\LRRK2I2020T.Scer\UAS Parkinson's Disease models; some aspects of the eye phenotype (black lesions) are suppressed, whereas others (such as bristle loss) are enhanced.

Detailed Description
Statement
Reference

The expression of Hsap\PINK1Scer\UAS.cYa under the control of Scer\GAL4Act.PU in midgut clones results in enterocytes during pupariation exhibiting a significant decrease in the relative mitochondrial content and a significant increase in the relative amount of autophagosomes, as assessed by a Atg8a fluorescent reporter, but no significant change in the transverse cell area, as compared to non-clone enterocytes.

Expression of Hsap\PINK1Scer\UAS.cYa under the control of Scer\GAL4GMR.PF at 29[o]C does not cause loss of pigmentation in the eye, but the eyes show some loss and disorganisation of interommatidial bristles.

Expression of Hsap\PINK1Scer\UAS.cYa, under the control of Scer\GAL4ple.PF has no effect on TH+ neuron number.

The increased relative amount of autophagosomes exhibited during pupariation by enterocytes in midgut clones expressing Hsap\PINK1Scer\UAS.cYa under the control of Scer\GAL4Act.PU is enhanced by the clonal co-expression of Hsap\PARK2Scer\UAS.cYa.

Co-expression of Hsap\PINK1Scer\UAS.cYa does not significantly suppress the loss of pigmentation in the eye, the roughness of the eye surface or the formation of holes in eye sections that are seen in flies expressing Hsap\LRRK2Scer\UAS.cVa under the control of Scer\GAL4GMR.PF at 29[o]C, but the formation of black lesions in the eye is partially suppressed. The loss of interommatidial bristles that is seen in flies expressing Hsap\PINK1Scer\UAS.cYa under the control of Scer\GAL4GMR.PF at 29[o]C is enhanced by co-expression of Hsap\LRRK2Scer\UAS.cVa.

Co-expression of Hsap\PINK1Scer\UAS.cYa does not significantly suppress the loss of pigmentation in the eye, the roughness of the eye surface or the formation of holes in eye sections that are seen in flies expressing Hsap\LRRK2I1122V.Scer\UAS under the control of Scer\GAL4GMR.PF at 29[o]C. The loss of interommatidial bristles that is seen in flies expressing Hsap\PINK1Scer\UAS.cYa under the control of Scer\GAL4GMR.PF at 29[o]C is enhanced by co-expression of Hsap\LRRK2I1122V.Scer\UAS.

Co-expression of Hsap\PINK1Scer\UAS.cYa does not significantly suppress the loss of pigmentation in the eye, the roughness of the eye surface or the formation of holes in eye sections that are seen in flies expressing Hsap\LRRK2I2020T.Scer\UAS under the control of Scer\GAL4GMR.PF at 29[o]C, but the formation of black lesions in the eye is partially suppressed.

Mitochondrial integrity in Pink1dsRNA.Scer\UAS (under the control of either Scer\GAL4da.G32 or Scer\GAL4Mhc.PW) mutants is restored through co-expression of Hsap\PINK1Scer\UAS.cYa.

Mitochondrial integrity in Pink1dsRNA.Scer\UAS (under the control of either Scer\GAL4da.G32 or Scer\GAL4Mhc.PW) mutants is not restored through co-expression of Hsap\PINK1ΔC.Scer\UAS.
