Scer\FLP1 mediated chromosome translocation using PBac{WH}Tkf03824 and P{XP}d08303 has resulted in deletion of Tk. The expression of a neighboring gene, mfas, is not affected.
Deletion resulting from Scer\FLP1 mediated chromosome translocation using PBac{WH}Tkf03824 and P{XP}d08303, which removes the coding region of Tk.
Homozygous TkΔ1 mutant flies exhibit reduced sensitivity to (S,Z,Z)-CH503 (a more potent stereoisomer of the gustatory sex pheromone CH503): in contrast to wild type they do not exhibit increased courtship defects in the presence of (S,Z,Z)-CH503 compared with controls.
TkΔ1/Df(3R)ΔTk2 mutant flies exhibit reduced sensitivity to (S,Z,Z)-CH503 (a more potent stereoisomer of the gustatory sex pheromone CH503): in contrast to wild type they do not exhibit increased courtship defects in the presence of (S,Z,Z)-CH503 compared with controls.
TkΔ1 mutant flies are fully viable and fertile and exhibit no obvious anatomical abnormalities of differences from controls in locomotor activity. Homozygotes show a significant decrease in male-male aggressive behavior compared to heterozygous controls. Males copulate with virgin females at efficiencies comparable to those observed for heterozygous control males. Copulation latency and latency are unaffected, as is the frequency of unilateral wing extension "singing" toward mated females. The frequency of unilateral wing extension toward other males is also not consistently different from controls.
TkΔ1/Df(3R)ΔTk2 transheterozygotes show a significant decrease in male-male aggressive behavior compared to heterozygous controls.
Df(3R)ΔTk2/TkΔ1 is a suppressor | partially of abnormal behavior | adult stage | heat sensitive phenotype of Scer\GAL4Tk, TrpA1UAS.(B).cKa
TkΔ1/Df(3R)ΔTk2 partially suppresses the aggression-promoting effect of activating neurons labeled with the P{Tk-GAL4.TH}5Fa insertion of Scer\GAL4Tk through expression of thermosensitive TrpA1Scer\UAS.(B).cKa.
Df(3R)ΔTk2/TkΔ1 is rescued by TkUAS.cAa/Scer\GAL4Tk
Df(3R)ΔTk2/TkΔ1 is rescued by TkUAS.cAa/Scer\GAL4dimm-929
Df(3R)ΔTk2/TkΔ1 is rescued by TkUAS.cAa/Scer\GAL4Tk
Df(3R)ΔTk2/TkΔ1 is rescued by TkUAS.cAa/Scer\GAL4GMR61H07
TkΔ1 is not rescued by TkUAS.cAa/Scer\GAL4NPF.1
TkΔ1 is not rescued by TkUAS.cAa/Scer\GAL4dimm-929
Df(3R)ΔTk2/TkΔ1 is not rescued by TkUAS.cAa/Scer\GAL4Tk
Expression of TkScer\UAS.cAa under the control of Scer\GAL4Tk rescues the reduced sensitivity to (S,Z,Z)-CH503 (a more potent stereoisomer of the gustatory sex pheromone CH503) seen in TkΔ1/Df(3R)ΔTk2 mutant flies. As in wild type, courtship defects are seen in these flies in response to (S,Z,Z)-CH503.
Expression of TkScer\UAS.cAa under the control of Scer\GAL4NPF.1 does not rescue the reduced sensitivity to (S,Z,Z)-CH503 (a more potent stereoisomer of the gustatory sex pheromone CH503) seen in homozygous TkΔ1 mutant flies. As in TkΔ1 mutants alone, the courtship defects observed in response to CH503 in wild type are not seen.
Expression of TkScer\UAS.cAa under the control of Scer\GAL4dimm-929 does not rescue the reduced sensitivity to (S,Z,Z)-CH503 (a more potent stereoisomer of the gustatory sex pheromone CH503) seen in homozygous TkΔ1 mutant flies. As in TkΔ1 mutants alone, the courtship defects observed in response to CH503 in wild type are not seen.
Expression of TkScer\UAS.cAa under the control of Scer\GAL4dimm-929 rescues the reduced sensitivity to (S,Z,Z)-CH503 (a more potent stereoisomer of the gustatory sex pheromone CH503) seen in TkΔ1/Df(3R)ΔTk2 mutant flies. As in wild type, courtship defects are seen in these flies in response to (S,Z,Z)-CH503.
Expression of TkScer\UAS.cAa under the control of Scer\GAL4Tk (either the P{Tk-GAL4.TH}5Fa or P{Tk-GAL4.TH}3Ma insertion) fully rescues the defects in aggressive behavior seen in TkΔ1/Df(3R)ΔTk2 mutant males. No rescue is seen when TkScer\UAS.cAa is driven by another insertion of the same construct, P{Tk-GAL4.TH}2Ma, or Scer\GAL4GMR61H07.
