The R23C12 enhancer fragment is fused upstream of the Drosophila synthetic core promoter (DSCP) followed by the GAL4(DBD)::Zip- hemidriver (component of the 'Split GAL4' system, contains the GAL4 DNA-binding domain). A Hsp70 transcriptional terminator is present.

Thermogenetic activation (using TrpA1UAS.cUa at 32[o]C) of neurons targeted by the split-GAL4 pair of Scer\GAL4DBD.R23C12 and Hsap\RELAAD.R76F05 significantly reduces carbon dioxide avoidance in starved adults, compared to controls; the effect is significantly smaller in fed flies. There is no reduction in carbon dioxide avoidance when neurons are activated in association with carbon dioxide exposure in advance of, and not during, preference testing.
Optogenetic activation (using Cnoc\ChR1Cs.IVS.20xUAS.Venus with 617 nm (red) light) of neurons targeted by the split-GAL4 pair of Scer\GAL4DBD.R23C12 and Hsap\RELAAD.R76F05 in adults results in a significant attraction to the illuminated area, no attraction is observed in controls.
Split-GAL4 transgene, constructed as described in FBrf0212052.
