Inhibition (using Hsap\KCNJ25XUAS.EGFP) of neurons targeted by the split-GAL4 pair of Scer\GAL4DBD.R95H02 and Hsap\RELAAD.R76F05 in third instar larvae significantly impairs short-term associative appetitive olfactory memory following training with fructose, but not low salt or aspartic acid, with no effect on innate odor (n-amylacetate) or fructose preference, compared to controls.

Optogenetic activation (using Crei\ChR2XXL.UAS or Crei\ChR2XXL.UAS.tdTomato with 470 nm (blue) light, but not using Cnoc\ChR1Cs.IVS.20xUAS.Venus with 630 nm (red) light) of neurons targeted by the split-GAL4 pair of Scer\GAL4DBD.R95H02 and Hsap\RELAAD.R76F05 in third instar larvae, in combination with a paired odor, results in a significant increase in short-term associative attractive olfactory memory, compared to controls.

Thermogenetic activation (using TrpA1UAS.cUa at 32[o]C) of neurons targeted by the split-GAL4 pair of Scer\GAL4DBD.R23C12 and Hsap\RELAAD.R76F05, or the split-GAL4 pair of Scer\GAL4DBD.R80G12 and Hsap\RELAAD.R76F05, significantly reduces carbon dioxide avoidance in starved adults, compared to controls; the effect is significantly smaller in fed flies. There is no reduction in carbon dioxide avoidance when Scer\GAL4DBD.R23C12 and Hsap\RELAAD.R76F05 neurons are activated in association with carbon dioxide exposure in advance of, and not during, preference testing.

Optogenetic activation (using Cnoc\ChR1Cs.IVS.20xUAS.Venus with 617 nm (red) light) of neurons targeted by the split-GAL4 pair of Scer\GAL4DBD.R23C12 and Hsap\RELAAD.R76F05, or the split-GAL4 pair of Scer\GAL4DBD.R80G12 and Hsap\RELAAD.R76F05, in adults results in an attraction to the illuminated area, which is significant in the case of Scer\GAL4DBD.R23C12 and Hsap\RELAAD.R76F05 neurons, no attraction is observed in controls.
