Description

Twenty miRNA binding sites with mismatches at positions 9-12 (creating a "Sponge", designed to sequester miRNAs and thus knocking-down endogenous miRNA activity) are placed in the 3'UTR of mCherry under the control of 10 Gal4 UAS binding sites.

Upon the expression of mir-8^{sponge.V2.UAS.mCherry} under the control of Scer\GAL4^Mef2.PR, adult muscles still form but there is a significant increase in the final number of persistent muscle precursors (i.e. Zfh1-positive cells) in indirect flight muscles, as compared to controls.

Scer\GAL4^tub.PU-mediated expression of mir-8^{sponge.V2.Scer\UAS.T:Disc\RFP-mCherry} results in a deformed leg phenotype.
