Ectopic expression of Hsap\MAPTScer\UAS.0N4R.T:Zzzz\FLAG under the control of Scer\GAL4ptc.PU results in an increased number of cells in mitosis in the third instar larval wing disc, however there is no corresponding increase in S-phase cell numbers, suggesting that the cell-cycle duration is not shorter compared to controls. Levels of cell death (detected by Caspase-3 staining) are also increased in the Hsap\MAPTScer\UAS.0N4R.T:Zzzz\FLAG expressing region of the wing disc and the size of the corresponding area of the adult wing - the intervein region between L3 and L4 veins - is significantly reduced (measured at the wing margin) compared to controls.

Expression of Hsap\MAPTScer\UAS.0N4R.T:Zzzz\FLAG under the control of any of the following Scer\GAL4ptc.PU, Scer\GAL4dpp.PU or Scer\GAL4Bx-MS1096 leads to mitotic spindle defects - monopolar spindle - and chromosomal poly- and aneuploidy in third instar larval wing disc cells. Cells expressing Hsap\MAPTScer\UAS.0N4R.T:Zzzz\FLAG (under the Scer\GAL4ptc.PU driver) show failure of the duplicated centrosomes to move apart to the opposite poles, thus leading to the monopolar mitotic spindle.

Expression of Hsap\MAPTScer\UAS.0N4R.T:Zzzz\FLAG under the control of Scer\GAL4GMR.PU leads to rough eye phenotype in the adults, expression of single copy of the transgene has a rather mild effect: missing bristles but almost no ommatidial disorganization, expression of two copies of Hsap\MAPTScer\UAS.0N4R.T:Zzzz\FLAG results in stronger phenotype with more bristles missing and ommatidial disorganization at the posterior side of the eye.

Expression of Hsap\MAPTScer\UAS.0N4R.T:Zzzz\FLAG under the control of Scer\GAL4insc-Mz1407 results in mitotic defects (monopolar spindle, chromosomal aneuploidy) in the brain neuroblasts of third instar larvae.

Co-expression of Hsap\MAPTCter.Scer\UAS.T:Zzzz\FLAG together with Hsap\MAPTScer\UAS.0N4R.T:Zzzz\FLAG under the control of Scer\GAL4GMR.PU results in rough eye phenotype (missing interommatidial bristles, ommatidial fusions) in the adults. The phenotype is the same as when two copies of full-length Hsap\MAPTScer\UAS.0N4R.T:Zzzz\FLAG are used.

The reduced size of the L3-L4 intervein region (measured at the wing margin) characteristic for adult flies expressing Hsap\MAPTScer\UAS.0N4R.T:Zzzz\FLAG under the control of Scer\GAL4ptc.PU is decreased further by combination with Klp61Furc-1 in heterozygous state.

The rough eye phenotype (missing interommatidial bristles) observed in adult flies expressing single copy of Hsap\MAPTScer\UAS.0N4R.T:Zzzz\FLAG under the control of Scer\GAL4GMR.PU is exacerbated further (more missing bristles, ommatidial fusion) by combination with Klp61Furc-1 in heterozygous state. The resulting phenotype is similar or even stronger than that seen in flies expressing two copies of Hsap\MAPTScer\UAS.0N4R.T:Zzzz\FLAG.

The rough eye phenotype (missing interommatidial bristles) characteristic for flies expressing Hsap\MAPTScer\UAS.0N4R.T:Zzzz\FLAG under the control of Scer\GAL4GMR.PU can be almost completely suppressed (hardly any bristles missing) by expression of Klp61FUbi-p63E.T:Avic\GFP.
