ScerGAL4Tmc.3.0 is expressed in a labellar multidendritic neuron in each of the two bilaterally symmetrical labella. The cell body of these neurons is adjacent to the I6 sensilla at the proximal part of the labella, while the dendritic branches innervate the sensilla, predominately, but not exclusively, the L-type sensilla.
Expression of ScerGAL4Tmc.3.0 in the adult antennal lobe projection neurons and Kenyon cells may be ectopic rather than representative of Tmc.
Inhibition (using Hsap\KCNJ2IVS.10xUAS.EGFP) of Scer\GAL4Tmc.3.0 cells in adult females significantly increases preference for softer 100 mM sucrose-containing, but not plain, agarose for egg laying, compared to controls; inhibition does not affect preference for 100 mM sucrose-containing (rather than plain) agarose for egg laying, compared to controls; increased preference for softer 100 mM sucrose-containing agarose is still seen when inhibition is restricted to a subset of Scer\GAL4Tmc.3.0 cells using Scer\GAL80VGlut.PV.
Inhibition (using Ctet\tetXTNT-LC.UAS) of Scer\GAL4Tmc.3.0 cells in adult females significantly increases preference for softer 100 mM sucrose-containing agarose for egg laying, compared to controls.
Optogenetic activation (using Cnoc\ChR1::Csub\ChRCsChrimson.IVS.20xUAS.Venus with red light) of a subset of Scer\GAL4Tmc.3.0 cells (restricted using Scer\GAL80VGlut.PV) in adult females significantly decreases egg laying, compared to baseline.
Thermogenetic activation (using TrpA1UAS.(B).cKa at 30[o]C) of Scer\GAL4Tmc.3.0 cells in adult females significantly decreases egg laying, compared to baseline.
