Expression of TFAMUAS.cCa under the control of Scer\GAL4tim.PE (alone or in combination with either Scer\GAL80Pdf.PS or tub-Gal80[ts], to restrict expression to the adult stage) but not Scer\GAL4Pdf.PU, leads to a significant reduction in the amplitude of locomotor activity rhythm (circadian rhythm) under constant darkness when compared to control adults.

Expression of TFAMUAS.cCa under the control of Scer\GAL4Toll-6-D42 leads to climbing and wing inflation defects when compared to controls.

Expression of TFAMUAS.cCa under the control of Scer\GAL4nSyb.PS leads to almost complete late pupal lethality.

Expression of TFAMUAS.cCa under the control of Scer\GAL4nSyb.PS in the TFAMc01716/+ heterozygous mutant background leads to climbing and wing inflation defects when compared to controls.

Expression of TFAMScer\UAS.cCa under the control of Scer\GAL4109(2)80 results in slight shortening of the dendritic arbor in the class IV ddaC neurons in third instar larvae.

Expression of TFAMScer\UAS.cCa under the control of Scer\GAL4da.PU is first instar larval lethal; expression under the Scer\GAL4VGlut-OK371 driver at 25[o]C is partially pupal lethal, the surviving adults do not live beyond a week and display severe climbing ability defects, at 29[o]C the expression is fully pupal lethal. Neither the number of motoneuron cell bodies nor the level of cell death in the ventral nerve cord is significantly affected in third instar larvae. However, the number of active zones in the neuromuscular junctions (NMJs) is significantly decreased (while the number of boutons is not changed) and the number and volume of presynaptic mitochondria at the NMJ is also strongly reduced relative to controls, although their ultrastructure appear normal. TFAMScer\UAS.cCa expression affects mitochondria in other parts of the motor neuron as well: the cell body mitochondria are less reticular with more punctate morphology and both the number and volume of mitochondria in the distal axon is reduced, no significant effect is observed in the proximal axon.

Expression under the Scer\GAL4Toll-6-D42 also reduces adult climbing ability and lifespan and leads to failed wing inflation in about 50% of the adult flies.

However, the adult locomotion and wing inflation defects of TFAMScer\UAS.cCa expressing flies as well as their reduced lifespan are significantly improved by co-expression of either simaHMS00833 or simaHMS00832. The loss of mitochondria at the larval neuromuscular junction induced by Scer\GAL4VGlut-OK371-driven expression of TFAMScer\UAS.cCa is not affected by co-expression of simaHMS00833, while the reduction in the number of active zones at the NMJ is fully rescued.
