Synj1/Synj2 transheterozygotes expressing SynjScer\UAS.cVa under the control of Scer\GAL4nSyb.PU present insignificant differences in adult lifespan under starvation conditions and in the number of intact ommatidia upon 7 day exposure to either constant light or constant darkness, as compared to controls; their third instar larval neuromuscular junctions do not present any significant defects, as shown by insignificant differences in the amplitude and frequency of miniature and spontaneous excitatory junctional currents under basal conditions, and by an apparently normal induction of mature autophagosomes (detected as fluorescent Atg8 puncta) in response to direct electric nerve stimulation, as compared to controls.

The expression of SynjScer\UAS.cVa under the control of Scer\GAL4nSyb.PU rescues the larval lethality of Synj1/Synj2 transheterozygotes; this expression also rescues the decreased synaptic transmission in electroretinograms measured in Synj1 homozygous eyes within mosaic adults, as compared to controls.

The expression of SynjScer\UAS.cVa under the control of Scer\GAL4Toll-6-D42 rescues the third instar larval neuromuscular junction defects of Synj1/Synj2 transheterozygotes, as it suppresses the failure to sustain neurotransmission under high stimulation conditions and suppresses all the following presynaptic defects: the increase number of satellite boutons, the decrease in endocytosis levels, the failure to starvation-induce mature autophagosomes (detected as fluorescent Atg8 puncta) and autolysosomes (detected as fluorescent Lamp1 puncta), and the concomitant decrease in synaptic vesicles density and increase in cisternae density at boutons under starvation conditions.
