Complete deletion of the DnaJ-1 coding sequence.
DnaJ-1A/DnaJ-1C transheterozygotes are partially lethal, since only a small minority reaches adulthood, but do not show obvious morphological defects. DnaJ-1C homozygous and DnaJ-1A/DnaJ-1C and DnaJ-1C/Df(3L)BSC884 transheterozygous third instar larvae present a significant increase in both the number and size of circulating crystal cells (identified as Lozenge-positive or PPO1-positive cells, which are still capable of melanization upon heat treatment), but circulating cells have a unaffected size, as compared to controls; heterozygotes also exhibit an increase in the size, but not number, of crystal cells, as compared to controls.
DnaJ-1C/DnaJ-1A has increased cell size | third instar larval stage phenotype, suppressible by lzUAS.cBa/Scer\GAL4lz-gal4
DnaJ-1C/DnaJ-1A has increased cell number | third instar larval stage phenotype, suppressible by lzUAS.cBa/Scer\GAL4lz-gal4
DnaJ-1C/DnaJ-1A has increased cell size | third instar larval stage phenotype, suppressible by N[+]/N55e11
DnaJ-1C/DnaJ-1A has increased cell number | third instar larval stage phenotype, suppressible by N[+]/N55e11
DnaJ-1C/DnaJ-1A has embryonic/larval crystal cell | third instar larval stage phenotype, suppressible by lzUAS.cBa/Scer\GAL4lz-gal4
DnaJ-1C/DnaJ-1A has embryonic/larval crystal cell | third instar larval stage phenotype, suppressible by N[+]/N55e11
The increased number and size of circulating crystal cells observed in DnaJ-1A/DnaJ-1C transheterozygous third instar larvae is suppressed by the expression of lzScer\UAS.cBa under the ontrol of Scer\GAL4lz-gal4.
DnaJ-1C/DnaJ-1A is rescued by DnaJ-1UAS.cKa/Scer\GAL4lz-gal4
DnaJ-1C/DnaJ-1A is not rescued by DnaJ-1ΔJ.UAS.Tag:FLAG/Scer\GAL4lz-gal4
The increased number and size of circulating crystal cells present in DnaJ-1A/DnaJ-1C transheterozygous third instar larvae are rescued by the expression of DnaJ-1Scer\UAS.cKa, but not of DnaJ-1ΔJ.Scer\UAS.T:Zzzz\FLAG, under the control of Scer\GAL4lz-gal4.
