Abi5/Df(3R)su(Hw)7 third instar larvae display NMJ overgrowth with supernumerary boutons (both mature and satellite), increased evoked excitatory junction potential amplitudes and uncoordinated motor behavior, with a faster roll-over than controls in a behavioral assay. A significant increase in the number of synaptic boutons containing microtubule loops points towards an increase in microtubule stability.

When expressing AbiUAS.Tag:HA under the control of Scer\GAL4Abi.PK in Abi5/Df(3R)su(Hw)7 third instar larvae, evoked excitatory junction potential amplitudes decrease significantly, below control levels.

Bouton numbers are unaffected in Abi5/+ larvae.

Expressing AbiUAS.Tag:HA under the control of either Scer\GAL4Abi.PK or Scer\GAL4da.G32 fully rescues pupal lethality of Abi5/Df(3R)su(Hw)7 flies, while expression under the control of Scer\GAL4elav-C155, Scer\GAL4how-24B or under the combined control of Scer\GAL4elav-C155 and Scer\GAL4how-24B results in only a partial rescue.

Expressing AbiUAS.Tag:HA under the control of Scer\GAL4elav-C155 (but not Scer\GAL4how-24B) rescues the supernumerary phenotype of both mature and satellite boutons at the NMNJ and the uncoordinated phenotype of Abi5/Df(3R)su(Hw)7 third instar larvae in a roll over assay.

The supernumerary phenotype of mature and satellite boutons at the NMNJ of Abi5/Df(3R)su(Hw)7 third instar larvae is rescued by expressing AbiW452K.UAS.Tag:HA or Abi4YE.UAS.Tag:HA, but not AbiΔ30-65.UAS.Tag:HA, AbiΔ123-175.UAS.Tag:HA or Abi4YF.UAS.Tag:HA under the control of Scer\GAL4elav-C155.
