Description

UASt regulatory sequences drive expression of a mutated form of smo in which the five putative PKA phosphorylation sites in the C-terminal, cytoplasmic tail have been mutated (amino acid replacements S667A, S687A, S740A, T755A and T785A). The smo protein is tagged with Tag:FLAG at the C-terminal end.

Expression of smo5A.Scer\UAS.T:Zzzz\FLAG under the control of Scer\GAL4C-765 results in a reduction in the distance between veins III and IV in the wing and a partial fusions of these veins proximal to the anterior crossvein. The reduction in distance between wing veins III and IV and partial fusions of these veins proximal to the anterior crossvein that is seen in flies expressing smo5A.Scer\UAS.T:Zzzz\FLAG under the control of Scer\GAL4C-765 are strongly enhanced by smo3/+.

The disrupted wing vein phenotype characteristic for flies expressing smo5A.Scer\UAS.T:Zzzz\FLAG under the control of Scer\GAL4C-765 is further exacerbated by combination with a single copy of any of Kdm4AKO or Kdm4BKO, ameliorated by combination with JHDM2KO and not significantly modified by heterozygosity of any of the following: Kdm2KO, UtxΔ, lid10424, Jarid2KO, NO66KO, JMJD5KO, JMJD7KO, Df(3R)HSPBAP1-KO or JMJD4KO. Combination with PSRFM1 shows a tendency towards worsening the phenotype but the effect is not statistically significant.

Co-expression of smo5A.Scer\UAS.T:Zzzz\FLAG significantly suppresses the withdrawal responses to a 38[o]C probe (without pre-exposure to UV) seen in early third instar larvae with TkR99DScer\UAS.T:Avic\GFP-EGFP over-expressed by Scer\GAL4ppk.1.9.

The reduction in distance between wing veins III and IV and partial fusions of these veins proximal to the anterior crossvein that is seen in flies expressing smo5A.Scer\UAS.T:Zzzz\FLAG under the control of Scer\GAL4C-765 are suppressed by ptcG12/+; the resulting flies have nearly normal wings.
