Detailed Description

100% of embryos derived from homozygous or gd2/Df(1)KA10 females have a class I phenotype (they are strongly dorsalised, lacking both ventral denticles and filzkorper).

In embryos generated from gd2 mothers, Tl is inactive, dl remains exclusively cytoplasmic and the embryos fail to form filzkorper and ventral denticles.

Injection of pll::torcGa into embryos from gd2 mothers results in the appearance of filzkorper and ventral denticle belts.

Cell intercalation in lateralized gd mutant embryos proceeds normally during germ band extension.

Transheterozygous combinations of gd mutations produce a range of cuticular phenotypes, ranging from cuticle only having dorsal characteristics, to cuticle having some ventral characteristics, such as ventral setal belts, depending on the alleles used. Some combinations of alleles complement each other.

75% of embryos derived from gd2/gd5 females have a class III phenotype; ventral cuticle is reduced, but some ventral abdominal setal belt material is retained, the anterior midgut invagination and ventral furrow are reduced or absent, gut formation is abnormal and the tracheal pits are located closer to the ventral midline than normal. 20% have a class II phenotype; embryos lack ventral cuticle, the filzkorper are present and increased amounts of cuticle show dorsal characteristics compared to wild-type. 36% of embryos derived from gd2/gd6 females hatch, 35% either do not complete head involution or develop into apparently normal L1 larvae which either fail to hatch or die soon after hatching, and 29% have a class III phenotype.

Embryonic defects detectable at gastrulation.

Homozygous females lay eggs that fail to hatch.

Injection of snkXa RNA does not rescue the dorsalised phenotype of embryos derived from gd2/gd2 females.

When piphs.PS is uniformly expressed in gd2 females, the resulting embryos are dorsalised.

The zygotic semi-lethality of cact7 homozygotes is not suppressed by gd2.

Approximate order of allelic severity, from weakest to strongest is: gd5 > gd1 > gd3 > gd6 > gd2 > gd4 > gd7 = gd8 = gd9 = gd10 = gd11 = gd12.

Blocks production of extracellular Tl ligand, consequently signal transduction is not initiated and dl remains cytoplasmic.
