UASt regulatory sequences drive expression of a dominant-negative form of btl. The encoded protein contains the signal peptide, extracellular and transmembrane domains, but only 100 amino acids of the cytoplasmic tail.

The expression of btlDN.Scer\UAS under the control of Scer\GAL4btl.PS results in tracheoblast cells not migrating into the leg discs and leads to only a few btl-positive neurons localizing in the eye disc and in the brain lobe, as compared to controls.

Expression of btlDN.Scer\UAS in tracheal progenitors and their descendants under the control of Scer\GAL4esg-p127 and Scer\GAL4Act5C.PI blocks migration and diminishes or eliminates the formation of pupal abdominal tracheae.

Scer\GAL4btl.PS-mediated expression of btlDN.Scer\UAS results in shorter tracheal ganglionic branches.

Expression of btlDN.Scer\UAS under the control of Scer\GAL4btl.PS from the late second larval instar stage onwards reduces or eliminates secondary branches and coiled tracheolar (CT) cells in the pupal tracheal system.

Embryos that express btlDN.Scer\UAS under the control of Scer\GAL4btl.PS do not show tracheal branch integrity defects but do show mild defects in fusion and terminal branching.

Expression of btlDN.Scer\UAS under the control of Scer\GAL4ato.3.6 does not affect initial extension of dorsal cluster neuron axons at 20%-30% pupal development but results in a large increase (from 11.7 to 18.9) in the number of dorsal cluster neuron axons that cross toward the medulla in adult brains.

The wing discs of third instar btlDN.Scer\UAS; Scer\GAL4btl.PS larvae have decreased numbers and migration of air sac tracheoblasts.

Expression of btlDN.Scer\UAS under the control of Scer\GAL4dpp.blk1 has no effect on furrow formation in the eye disc.

btlDN.Scer\UAS, when driven by Scer\GAL4A9, leads to significantly lower numbers of GB tracheal branches than seen in wild-type. btlDN.Scer\UAS when driven by Scer\GAL4bs-23.26 leads to significantly lower numbers of LG tracheal branches than seen in wild-type.

Scer\GAL4hs.2sev-mediated expression of btlDN.Scer\UAS 4 hours AEL exerts an inhibitory effect on tracheal and midline glia (MGP) cell migration. Heat shock at different stages of tracheal migration reveals btl is obligatory primarily for the onset of the tracheal migration process. btl is also required for the formation of tracheoles; no tracheoles form in heat treated flies.

All larvae co-expressing psqKK111691 and EgfrUAS.cBa under the control of Scer\GAL4ap-md544 during larval stages (using Gal80[ts] for the temporal regulation of expression) wing discs tumors, associated with higher numbers of myoblasts. These phenotypes are partially suppressed by the co-expression of btlDN.UAS.

Scer\GAL4btl.PS-mediated expression of btlDN.Scer\UAS suppresses the tracheal cyst phenotype of exp135 embryos.

Coexpression of Rac1Scer\UAS.cLa and btlDN.Scer\UAS, under the control of Scer\GAL4ato.3.6, results in the dominance of the Rac1Scer\UAS.cLa phenotype, with a reduction in the number of dorsal cluster neuron axons crossing toward the medulla.

Coexpression of btlDN.Scer\UAS and Rac1N17.Scer\UAS, under the control of Scer\GAL4ato.3.6, results in all dorsal cluster neuron axons crossing the optic chiasm, instead of the majority retracting and branching within the lobula. This indicates that the Rac1N17.Scer\UAS phenotype is dominant.

Expression of btlDN.Scer\UAS under the control of Scer\GAL4ato.3.6 in a dsh1/+ background suppresses the reduction of dorsal cluster neuron axon extension phenotype seen in either single mutant, with the number of dorsal cluster neuron axons crossing the optic chiasm restored to almost wild-type levels.
