Description

UASt regulatory sequences drive expression of a wild-type csw coding region with a Tag:Myr(Src64B) myristoylation sequence fused to its N-terminus. The myristoylation signal results in the protein being constitutively targetted to the plasma membrane, producing a gain-of-function protein.

Expression of csw::Src64Bsrc90.Scer\UAS under the control of Scer\GAL4btl.PS results in the presence of extra terminal cells in the larval tracheal system.

Expression of csw::Src64Bsrc90.Scer\UAS under the control of Scer\GAL4CY2 results in dorsalised eggshells. Expression of csw::Src64Bsrc90.Scer\UAS under the control of Scer\GAL455B results in dorsalised embryos. Expression of csw::Src64Bsrc90.Scer\UAS under the control of Scer\GAL471B results in ectopic wing veins. Expression of csw::Src64Bsrc90.Scer\UAS under the control of Scer\GAL4twi.PG results in an increase in the number of eve-positive mesodermal cells in the embryo, most likely due to duplication of the DA1 muscle precursors. Expression of two copies of csw::Src64Bsrc90.Scer\UAS under the control of Scer\GAL4twi.PG results the formation of eve-positive cells outside of their normal boundary. Embryos expressing csw::Src64Bsrc90.Scer\UAS under the control of Scer\GAL4twi.PG have the normal number of VA2 muscle precursor cells.

Expression driven by various Scer\GAL4 lines causes expansion of ventral ectodermal cell fates, aberrant tracheal cell migration, ectopic tracheal tissue, and increased numbers of select muscle and heart precursor cells.

Expression of csw::Src64Bsrc90.Scer\UAS under the control of Scer\GAL4twi.PG partially suppresses the loss of VA2 muscle precursor cells seen in csw5/Y embryos derived from homozygous female germline clones. The eve-positive muscle progenitors and subsequent pericardial and DA1 precursor cells are recovered in csw5/Y embryos that are also expressing csw::Src64Bsrc90.Scer\UAS under the control of Scer\GAL4twi.PG. Expression of csw::Src64Bsrc90.Scer\UAS under the control of Scer\GAL4twi.PG nearly completely suppresses the loss of VA2 muscle precursor cells seen in cswLE120/Y embryos derived from homozygous female germline clones (94% of hemisegments have VA2 cells). Expression of csw::Src64Bsrc90.Scer\UAS under the control of Scer\GAL4twi.PG significantly suppresses the loss of VA2 muscle precursor cells seen in spi3 embryos (67% of hemisegments have VA2 cells). Expression of csw::Src64Bsrc90.Scer\UAS under the control of Scer\GAL4twi.PG significantly suppresses the loss of VA2 muscle precursor cells seen in rhoPΔ38 embryos (69% of hemisegments have VA2 cells). Co-expression of csw::Src64Bsrc90.Scer\UAS significantly suppresses the loss of VA2 muscle precursor cells seen in embryos expressing EgfrDN.Scer\UAS.cBa under the control of Scer\GAL4twi.PG (71% of hemisegments have VA2 cells). Expression of csw::Src64Bsrc90.Scer\UAS under the control of Scer\GAL4twi.PG significantly suppresses the loss of VA2 muscle precursor cells seen in dosR31 embryos (41% of hemisegments have VA2 cells). Expression of csw::Src64Bsrc90.Scer\UAS under the control of Scer\GAL4twi.PG partially suppresses the loss of VA2 muscle precursor cells seen in Ras85DΔC40B embryos (21% of hemisegments have VA2 cells). Expression of csw::Src64Bsrc90.Scer\UAS under the control of Scer\GAL4twi.PG partially suppresses the loss of VA2 muscle precursor cells seen in phl11/Y embryos (10.5% of hemisegments have VA2 cells). Expression of csw::Src64Bsrc90.Scer\UAS under the control of Scer\GAL4twi.PG partially suppresses the loss of VA2 muscle precursor cells seen in ksrs628 embryos (14% of hemisegments have VA2 cells). msk5 dominantly suppresses the formation of extra eve-positive mesodermal cells seen in embryos expressing csw::Src64Bsrc90.Scer\UAS under the control of Scer\GAL4twi.PG. Expression of csw::Src64Bsrc90.Scer\UAS under the control of Scer\GAL4twi.PG does not affect the loss of VA2 muscle precursor cells seen in pntΔ88 embryos (82% of hemisegments have VA2 cells).
