Description

14.4kb BamHI-EcoRI genomic fragment that encompasses brm. The C-terminal two residues of brm have been replaced by sequence encoding the Tag:HA epitope followed by six His residues (details from FBrf0083116). A K804R mutation has been introduced into the brm coding sequence; this mutation is within the ATP-binding site and results in a dominant-negative protein.

Flies carrying one or two copies of brmK804R.T:Ivir\HA1 are viable. Flies carrying two copies of brmK804R.T:Ivir\HA1 in a wild-type background frequently show partial transformation of haltere to wing, as shown by an increase in haltere size and the appearance of ectopic bristles on the capitellum. This phenotype is also seen in Df(3L)th102/+ animals carrying two copies of brmK804R.T:Ivir\HA1. brm2/Df(3L)th102 animals carrying two copies of brmK804R.T:Ivir\HA1 do not survive to adulthood.

Female brm deficiency heterozygotes carrying an X-linked copy of brmK804R.T:Ivir\HA1 survive to adulthood, but males of this genotype are not viable.
