gclrev390 homozygotes and gclrev390/gclE100K transheterozygotes present severe decreases in the number of primordial germ cells in the early embryo (i.e. mitotic cycles 12/13), as compared to wild-type and heterozygous controls; gclrev390 heterozygotes do not display any obvious dominant defects in fertility, as shown by the insignificant differences in progression through oogenesis and in laid eggs, as compared to controls.
In gclrev390 embryos, the normal transcriptional quiescence seen at nuclear cycle 10 is not seen.
When transformed with the cmp44E rescue construct cmp44E+t8.5, flies homozygous for Df(2R)cmp44E-rev390 lack any visible phenotypes. Mothers homozygous for Df(2R)cmp44E-rev390, with one copy of cmp44E+t8.5, produce embryos that hatch normally. At the blastoderm stage, however, about half lack any germ cells and the remainder have fewer pole cells than control embryos. At stage 14 about 2/3 of embryos lack pole cells and the pole cell survival rate is much lower than in controls - only about 2/5 of pole cells formed at the blastoderm stage successfully reach the embryonic gonad. The proportion of fertile adults match the proportion of embryos that have at least one pole cell at stage 14. The loss of pole cells appears to occur at Stage 10. Examination of earlier stages reveals that 1/5 embryos fail to form detectable pole buds.
