In ruPLLb mutant third instar larval eye disc clones (generated using the eyFLP system) the general program of photoreceptor differentiation appears to be unperturbed and axons are correctly targeted to the lamina. However defects are seen in lamina cartridge neuron differentiation. Electroretinogram recordings from adult flies show that the eyes of ruPLLb mutants are able to properly depolarize in response to light. The 'on/off transients" (representing the activity of post-synaptic lamina cartridge neurons) that are normally seen in wild type are absent from the ECG.

In stage 16 ruPLLb mutant embryos, a significant number of ganglionic branch trachea fail to turn posteriorly and dorsally at the ventral midline and instead cross the midline or remain lingering on it. Associated glia are unaffected.

In ruPLLb flies, 84% of ommatidia have reduced numbers of photoreceptor cells and those with correct numbers show rotation defects.

ruPLLb mutant flies have malformed eyes, with collapsed and blistered ommatidia.

The regular array of facets in the eyes of homozygotes produces a rough eye phenotype. There is variable loss of photoreceptors in these mutant eyes, however few defects if any are seen in the recruitment of photoreceptors. Cone cells are dramatically under-recruited, and some pigment cells are seen to be missing.

Expression of rhoScer\UAS.T:Avic\GFP-EGFP in the R2, R5 and R8 photoreceptors under the control of Scer\GAL4MT14 fails to suppress the lamina cartridge neuron differentiation defects seen in the eye discs of ruPLLb mutant third instar larvae.

Expression of rho::ruR3L1-R1.Scer\UAS.T:Avic\GFP-EGFP (rho targeted to the ER) in the R2, R5 and R8 photoreceptors under the control of Scer\GAL4MT14 suppresses the lamina cartridge neuron differentiation defects seen in the eye discs of ruPLLb mutant third instar larvae.

Expression of rho::ruR1L1-R3.Scer\UAS.T:Avic\GFP-EGFP (ru that is not targeted to the ER) in the R2, R5 and R8 photoreceptors under the control of Scer\GAL4MT14 fails to suppress the lamina cartridge neuron differentiation defects seen in the eye discs of ruPLLb mutant third instar larvae.

One copy of Syx5AR113 partially rescues the lamina cartridge neuron differentiation defects seen in the eye discs of ruPLLb mutant third instar larvae. Neuronal markers are detected in a small population of cells at the posterior of the lamina.

Expression of rhoScer\UAS.T:Avic\GFP,T:Uuuu\KDEL in the R2, R5 and R8 photoreceptors under the control of Scer\GAL4MT14 fails to suppress the lamina cartridge neuron differentiation defects seen in the eye discs of ruPLLb mutant third instar larvae.

Expression of spic.Scer\UAS.T:Arus\HRP,T:SS-wg in the R8 photoreceptors under the control of Scer\GAL4MT14 fails to suppress the lamina cartridge neuron differentiation defects seen in the eye discs of ruPLLb mutant third instar larvae.

Expression of spic.Scer\UAS.T:Ivir\HA1 in the R8 photoreceptors under the control of Scer\GAL4MT14 fails to suppress the lamina cartridge neuron differentiation defects seen in the eye discs of ruPLLb mutant third instar larvae.

Mutants homozygous or heterozygous for the ruPLLb allele have enlarged cardiac chambers.

The effects of sliGA20 and ruPLLb on migration of ganglionic branch trachea approaching the ventral midline during stage 16 appear to be additive.

The eye phenotypes seen in ruPLLb homozygotes are partially rescued by sl9/sl9 : Most ommatidia have normal numbers of photoreceptor cells, but 50% show rotation defects.

Expression of ruScer\UAS.T:Avic\GFP-EGFP in all photoreceptors (R1 - R8) under the control of Scer\GAL4GMR.PF fully rescues the lamina cartridge neuron differentiation defects seen in the eye discs of ruPLLb mutant third instar larvae.

Expression of ruScer\UAS.T:Avic\GFP-EGFP in the R2, R5 and R8 photoreceptors under the control of Scer\GAL4MT14 fully rescues the lamina cartridge neuron differentiation defects seen in the eye discs of ruPLLb mutant third instar larvae.

Expression of ruScer\UAS.T:Avic\GFP-EGFP in the R1, R6 and R7 photoreceptors under the control of Scer\GAL4lz-gal4 partially rescues the lamina cartridge neuron differentiation defects seen in the eye discs of ruPLLb mutant third instar larvae. 20% of the wild type number of lamina cells express differentiation markers.

Expression of ruScer\UAS.T:Avic\GFP-EGFP in the R3, R4 and R7 photoreceptors under the control of Scer\GAL4hs.2sev fails to rescue the lamina cartridge neuron differentiation defects seen in the eye discs of ruPLLb mutant third instar larvae.
