The hatching rate of embryos from parents homozygous for marsEP2477 is 67%. Viability is also temperature-dependent and the hatching rate is further reduced to 39.2% at 28.5[o]C.

Gynandromorphs form approximately 0.05% of the progeny from marsEP2477 parents.

In metaphase marsEP2477 embryos, the length and the density of mitotic spindles is significantly shorter and less, respectively, than in controls. At anaphase and at telophase, neither the length nor the density of mitotic spindles are substantially affected, indicating that polymerisation of the mitotic spindles in marsEP2477 embryos is delayed, but not entirely prevented. Consistent with this, progression of mitotic events in patches of nuclei, each of which contain at leat two nuclei, is obviously delayed compared to those in adjacent nuclei in 13% of marsEP2477 embryos.

The frequencies of non-disjunction in male progeny from marsEP2477 homozygotes at 24[o]C and 28.5[o]C were 0.6% and 1.1% respectively, which are much higher than the gynandromorphy rate in the mars parent.

marsEP2477 overexpression in the eye disc under the control of Scer\GAL4GMR.PF results in cells in domain I being retained at interphase and many cells in domains II and III being retained at M-phase. This should be compared to the fact that most M-phase cells appear in domain I in control eye discs.

Eggs laid by marsEP2477 and mars1/marsEP2477 flies show a greatly reduced ability to hatch. Sperm tails can be visualised in early-arrested embryos laid by mutant females, indicating that sterility is caused by arrest in embryogenesis after fertilization.

Approximately 51% of embryos laid by marsEP2477 mothers fail to develop beyond embryogenesis. Many show terminal phenotypes at or shortly after gastrulation.

The hatching rate from tlk27-9/+; marsEP2477 females is 48%, a further reduction from the 67% for marsEP2477 embryos, indicating that mars genetically interacts with tlk.

The frequencies of non-disjunction in male progenies from tlk27-9/+; marsEP2477 and tlkΔ14/+; marsEP2477 females at 28.5[o]C is 6.1% and 4.4%; these rates are much higher than those from marsEP2477 (1.1%) or tlkΔ14/+ females (0.4%). This increase in frequency of non-disjunction is also temperature dependent, since there is no substantial increases in progeny from tlk27-9/+; marsEP2477 and tlkΔ14/+; marsEP2477 females at 24[o]C when compared to marsEP2477 or tlkΔ14/+ females.

All tlkΔ14/+; marsEP2477 embryos at either metaphase or anaphase exhibit asynchrony so severe that it is impossible to distinguish what phase an embryo belongs to. Asynchronous chromosome segregation is observed in embryos where most of the nuclei are likely to be at anaphase. Despite this severe asynchrony during chromosome congression or segregation, the morphology of the mitotic spindles in tlkΔ14/+; marsEP2477 embryos is not significantly different from that in marsEP2477 embryos.

Co-expression of marsEP2477 and tlkScer\UAS.cCa in the eye disc under the control of Scer\GAL4GMR.PF results in the number of M-phase cells in domain III being reduced to a level close to that of wild-type, showing that metaphase arrest induced by marsEP2477 overexpression is suppressed by tlkScer\UAS.cCa overexpression.

The lethality of embryos laid by mars1/marsEP2477 mothers is rescued by taccS863L.Ubi-p63E.T:Avic\GFP but not taccUbi-p63E.T:Avic\GFP expression.
