dgo380 testes have significantly fewer normal individualization complexes (ICs) while they have a similar number of abnormal ICs when compared to controls.

dgoEGFP.N, dgo380 individuals exhibit normal wings with normal trichome polarity.

The wings of homozygous dgo380 mutant flies exhibit strong polarity defects and display a stereotypical whorl phenotype.

Homozygous dgo380 mutant flies exhibit strong polarity defects in the eye, including loss of chirality and misoriented facets.

Overall length of the tracheal dorsal trunk is normal in hemizygous stage 16 embryos.

Many dgo380 ommatidia mis-rotate; the variance (s.d.) in degree of rotation is greater than in wild type in adult eyes.

Homozygotes show defects in ommatidial polarity.

The formation of holes in wings due to temperature shift of shi1/shi1 animals during pupal stages is moderately enhanced by dgo380/+.

In developing eyes which are mosaic for dgo380, when the R3 photoreceptor precursor is wild type, the ommatidial cluster adopts the correct polarity, but when the R3 precursor is dgo380, clusters often adopt random polarity, leading to symmetrical clusters or chirality flips.

The ommatidia of dgo380 mutant flies have either randomized chirality, are misrotated or are symmetrical (R3/R3 or R4/R4 cell pairs replace R3/R4 pairs).

dgo380 homozygotes have moderately severe planar polarity defects in wing hairs and ommatidia but only mild planar polarity defects in bristles. The eye defects include misrotation and inverted chirality (average 38.4+/-11.2 % of ommatidia) and symmetrical ommatidia (average 28.6+/-13.1 % of ommatidia). dgo380/Df(2R)27 flies appear to be phenotypically identical.

Vangstbm-6, dgo380, pkpk-sple-13 and dsh3, Vangstbm-6, dgo380, pkpk-sple-13 triple and quadruple heterozygotes exhibit mild wing trichome orientation defects in proximal regions.

dgo380/+ causes wing hair whirls in 44% of flies expressing Mks1GD9100 under the control of Scer\GAL4en-e16E.

dgo380/+ causes wing hair whirls in 89% of flies expressing CG15283GD13108 under the control of Scer\GAL4en-e16E. The enhancement of the CG15283GD13108 planar cell polarity (PCP)-phenotype by dgo380 is particularly robust.

ed1X5 dominantly enhances the phenotype of mis-rotation of ommatidia that is seen in dgo380 animals.

The ommatidial polarity defects caused by dgo380 are suppressed by one copy of bdgGMREP or bdg164.

The ommatidial polarity defects seen in bdgGMREP homozygotes are dominantly suppressed by dgo380.

dgo380 Vangstbm-6/Vangstbm-6 clones in the pupal wing (32 hours after puparium formation) cause neighbouring cells to point their trichomes away from the clone, as occurs with Vangstbm-6/Vangstbm-6 single mutant clones in the pupal wing.

dgo380 pkpk-sple-13/pkpk-sple-13 clones in the pupal wing (32 hours after puparium formation) do not affect polarity of trichomes in surrounding wild-type tissue.

Clones expressing fzScer\UAS.T:Avic\GFP-EGFP under the control of Scer\GAL4Act5C.PI in a pkpk-sple-13/pkpk-sple-13 background result in non-clonal cells pointing their trichomes away from the clone. This effect extends 8-9 cells from the clone boundary. This effect is still seen if the clones are also mutant for dgo380, but in this case the effect only extends 5-6 cells from the clone boundary.

Both a dgo380/+ and a Vangstbm-6/+ background enhances the ommatidial rotation phenotype of Scer\GAL4hs.2sev>shgdCR3h.Scer\UAS.T:Avic\GFP-rs eyes; a dgo380/+, Vangstbm-6/+ double mutant background further enhances this phenotype.

The proportion of symmetrical (R3/R3) ommatidia in the eyes of stanScer\UAS.cUa; Scer\GAL4sev.PM181 flies is significantly enhanced by heterozygosity for dgo380.

Expression of Mmus\Ankrd6Scer\UAS.cJa in the posterior compartment of the wing under the control of Scer\GAL4hh-Gal4 completely rescues the whorl phenotype seen in the wing bristles of dgo380 mutant flies. The whorl phenotype is still observed in the anterior compartment. Expression of Mmus\Ankrd6Scer\UAS.cJa under the control of Scer\GAL4hh-Gal4 also rescues the dgo380 eye polarity phenotypes.
