Description

P{SUPor-P}S2PKG08356 insertion disrupts the open reading frame of the S2P transcript at codon 261. An in-frame stop codon arises from P-element sequences four codons after the insertion site.

Viability is reduced in S2PKG08356/S2P1 mutant flies from S2PKG08356/S2PKG08356 homozygous mothers mated to S2P1 males. Viability is restored by feeding the flies medium enriched with fatty acids.

S2PKG08356/S2P1 larvae are comparable in size to control larvae 48 hours after egg laying. However, from this age mutant larvae are distinctly smaller than control larvae, and emerge on average 2 days late (or even later if animals are raised in conditions of crowding). By adulthood, mutants display a greater variability in body size compared to controls. Mutants show normal morphology.

Viability is reduced in S2PKG08356/S2P1 mutant flies from S2PKG08356/S2PKG08356 homozygous mothers mated to S2P1 males, compared to progeny from heterozygous mutant parents. This reduced viability is restored by feeding the flies on a fatty-acid enriched culture medium.

S2PKG08356/S2P1, IceΔ1/+ and S2PKG08356/S2P1, IceΔ1/IceΔ1 mutant larvae exhibit lethality between second and third instar stages. Lethality is rescued partially by supplementing fly food with fatty acids, but viability is still reduced compared to S2PKG08356/S2P1 mutants.

S2PKG08356/S2P1, Nc51/+ and S2PKG08356/S2P1, Nc51/Nc51 mutant larvae exhibit lethality between second and third instar stages. Lethality is rescued partially by supplementing fly food with fatty acids, but viability is still reduced compared to S2PKG08356/S2P1 mutants.

On unsupplemented medium, S2PKG08356/S2P1 mutants in the presence of two copies of HLH106D386A and carrying HLH106189/+, survive at less than 25% of the expected rate. When these animals are homozygous for HLH106189, survival at only 10% of the expected rate is observed. Survival is restored by supplementing the culture medium with free fatty acids.
