Eye and wing phenotypes (small rough eyes, abnormal wings) and some degree of lethality are seen in various PDZ-GEF allelic combinations, with their relative strength in the following order: PDZ-GEF4/PDZ-GEF6 = PDZ-GEF4/PDZ-GEF5 > PDZ-GEF3/PDZ-GEF4 > PDZ-GEFk13720/PDZ-GEF3 = PDZ-GEFk13720/PDZ-GEF4 > PDZ-GEFEP388/PDZ-GEF3 > PDZ-GEFk13720/PDZ-GEFk13720 > PDZ-GEFk13720/PDZ-GEFEP388 > PDZ-GEFEP388/PDZ-GEFEP388.
Many mutant females have three spermathecae instead of two, with different allelic combinations having different percentages of females with three spermathecae: PDZ-GEF3/PDZ-GEF3 22%, PDZ-GEF3/PDZ-GEF4 30%, PDZ-GEF4/PDZ-GEF5 59%, PDZ-GEF4/PDZ-GEF6 62%, PDZ-GEFk13720/PDZ-GEF4 % (all PDZ-GEFk13720/PDZ-GEF4, PDZ-GEFk13720/PDZ-GEF3 or PDZ-GEFEP388/PDZ-GEF3 females have two spermathecae).
The number of male germline stem cells is dramatically reduced in the testes of Gef263/Gef264 animals compared to wild-type. In Gef264/Gef265 or Gef264/Gef266 the reduction is even more dramatic, with some testes lacking germ cells altogether. The secondary spermatogonia (identifiable by their branched fusomes) move to the tip.
In Gef263/Gef264 testes, the hub is not lost, but expression of some marker genes in the hub is. The number of cyst progenitor cells is also reduced in these testes.
During larval stages, male germline stem cells in Gef263/Gef264 animals become detached from the hub. Marker localisation data suggests that the adherens junctions between the hub cells and the surrounding stem cells are impaired.

RV12.Scer\UAS.T:Hsap\MYC; Scer\GAL4upd1.PU rescues the formation of adherens junctions between male germline stem cells and hub cells in the germinal proliferation center of PDZ-GEF4/PDZ-GEF6 animals. The same phenotype in PDZ-GEF4/PDZ-GEF3 animals is rescued by shgScer\UAS.T:Avic\GFP-rs; Scer\GAL4upd1.PU.

PDZ-GEF5/PDZ-GEF4 is partially rescued by PDZ-GEF+t11.6
PDZ-GEF+t11.6 rescues eye and wing phenotypes of PDZ-GEF3/PDZ-GEF4, PDZ-GEF4/PDZ-GEF5 or PDZ-GEF4/PDZ-GEF6 flies. PDZ-GEF+t11.6 partially rescues the supernumerary spermathecae phenotype in PDZ-GEF4/PDZ-GEF5 flies.
Germline stem cell loss in the testes of Gef264/Gef266 animals is rescued by Gef26+t11.6.
