PDZ-GEF3/PDZ-GEF3 escapers have small rough eyes and abnormal wings. PDZ-GEF3/PDZ-GEF3 homozygous escapers or homozygous clones have ommatidia that lack various photoreceptors cells and display planar polarity defects. PDZ-GEF3/PDZ-GEF3 mutant wings often form ectopic veins in the middle inter-vein region or are fused with normal veins; wings may also have clumps on the surface (may be extra chaetae or sensilla).

Eye and wing phenotypes (small rough eyes, abnormal wings) and some degree of lethality are seen in various PDZ-GEF allelic combinations, with their relative strength in the following order: PDZ-GEF4/PDZ-GEF6 = PDZ-GEF4/PDZ-GEF5 > PDZ-GEF3/PDZ-GEF4 > PDZ-GEFk13720/PDZ-GEF3 = PDZ-GEFk13720/PDZ-GEF4 > PDZ-GEFEP388/PDZ-GEF3 > PDZ-GEFk13720/PDZ-GEFk13720 > PDZ-GEFk13720/PDZ-GEFEP388 > PDZ-GEFEP388/PDZ-GEFEP388.

Many mutant females have three spermathecae instead of two, with different allelic combinations having different percentages of females with three spermathecae: PDZ-GEF3/PDZ-GEF3 22%, PDZ-GEF3/PDZ-GEF4 30%, PDZ-GEF4/PDZ-GEF5 59%, PDZ-GEF4/PDZ-GEF6 62%, PDZ-GEFk13720/PDZ-GEF4 % (all PDZ-GEFk13720/PDZ-GEF4, PDZ-GEFk13720/PDZ-GEF3 or PDZ-GEFEP388/PDZ-GEF3 females have two spermathecae).

Gef263/Gef263 is semi-lethal. Escapers have small, rough eyes and abnormal wings.

The number of male germline stem cells is dramatically reduced in the testes of Gef263/Gef264 animals compared to wild-type. The hub is not lost, but expression of some marker genes in the hub is. The number of cyst progenitor cells is also reduced in these testes.

The testes of Gef263/Gef26k13720 animals have normal numbers of germline stem cells.

Gef263 homozygous germline stem cell clones develop normally.

During larval stages, male germline stem cells in Gef263/Gef264 animals become detached from the hub. Marker localisation data suggests that the adherens junctions between the hub cells and the surrounding stem cells are impaired.

PDZ-GEFk13720/PDZ-GEF3 females have two spermathecae, but with the introduction of Rap1rvB1/+ or Rap2lSH0581/+ extra spermathecal capsule materials are observed in some females; PDZ-GEFk13720/PDZ-GEF3 wing and eye phenotypes are also enhanced.

Expression of shgScer\UAS.T:Avic\GFP-rs driven by Scer\GAL4da.PU almost fully suppresses the supernumerary spermathecae phenotype (as well as wing and eye phenotypes) in PDZ-GEF4/PDZ-GEF3 flies.

The testes of PDZ-GEF3/PDZ-GEFk13720 animals have normal numbers of germline stem cells but numbers are reduced significantly if the animals are also Rap2l1/+ or RrvB1/+.

The reduction in germline stem cells seen in the testes of PDZ-GEF3/PDZ-GEF4 animals is significantly suppressed by Egfrf24/+.

shgScer\UAS.T:Avic\GFP-rs; Scer\GAL4upd1.PU rescues the formation of adherens junctions between male germline stem cells and hub cells in the germinal proliferation center of PDZ-GEF4/PDZ-GEF3.
