loqsPA.T:Zzzz\FLAG restores the embryonic viability of loqsKO mutants. The rescued males are fertile, however, females lay far fewer eggs than wild-type controls. None of the eggs produced by loqsKO mutants rescued by loqsPA.T:Zzzz\FLAG hatch, and few of the eggs display normal dorsal appendages.

loqsKO mutant females rescued by loqsPA.T:Zzzz\FLAG together with loqsPD.T:Zzzz\FLAG lay far fewer eggs than wild-type controls. None of the eggs produced by the rescued females hatch. Few of the eggs laid by the rescued females display normal appendages.

loqsKO mutant females rescued by either loqsPA.T:Zzzz\FLAG or the combination of loqsPA.T:Zzzz\FLAG and loqsPD.T:Zzzz\FLAG display fewer germline stem cells (GSCs) per germaria than wild-type controls.

The number of germline stem cells (GSCs) in loqsKO females rescued by all genotypes expressing loqsPB.T:Zzzz\FLAG (loqsPA.T:Zzzz\FLAG together with loqsPB.T:Zzzz\FLAG, or loqsPB.T:Zzzz\FLAG together with loqsPD.T:Zzzz\FLAG, or loqsPA.T:Zzzz\FLAG together with both loqsPB.T:Zzzz\FLAG and loqsPD.T:Zzzz\FLAG) appears normal compared to wild-type.

loqsKO mutants display a reduction in dsRNA processing and a subsequent ~60% reduction in siRNA levels.

loqsKO/+ does not significantly suppress the position effect variegation of the Sb1 allele that is seen in the T(2;3)SbV chromosome.

Homozygous embryos hatch and develop at rates similar to that of wild-type controls and most homozygous larvae undergo pupation. However, viability drops precipitously at the pupa/adult transition; most homozygotes die during eclosion, while approximately 10% emerge but are nearly paralysed and die shortly after.

Homozygous embryos derived from homozygous female germline clones do not develop past the first larval instar stage.

Newly emerged homozygous adult female escapers usually have empty germaria accompanied by one or two terminal stage egg chambers.

loqsf00791/loqsKO females have an average of 0.5 germline stem cells (GSCs) per germarium at 10 days after eclosion, compared to an average of 2-3 GSCs per germarium in wild-type females at this age. Differentiated cystoblasts with posterior-positioned fusomes are often seen at the normal location of GSCs in the germaria of loqsf00791/loqsKO females.

Homozygous female germline stem cell clones have a half-life of only 6 days, compared to a half-life of 21 days for control clones.
