Brca256E/Brca2KO third instar larval brain neuroblasts show a small increase in the frequency of chromosomal aberrations, as compared to controls.

Brca2KO homozygous females crossed to control males lay significantly less eggs than control females; the eggs show abnormal morphology, including egg-shell transparency, round or flattened shape, and fused or no dorsal appendages. Control females crossed to Brca2KO homozygous males do not show defects in egg production. Intercross of Brca2KO/+ males and females has no effect on the numbers of eggs or progeny.

Brca2KO homozygous females show ovary defects, from underdeveloped ovaries (fewer ovarioles, with small, disordered, and misshapen egg chambers, and few mature eggs) to complete ovarian dysgenesis; there is extensive disintegration of egg chambers starting at stage 7, including destruction of cell envelopes, DNA condensation, and extensive apoptosis. Brca2KO heterozygous females do not show these defects.

In Brca2KO homozygous males, nearly all testes are moderately-to-severely small and underdeveloped, and lack spermatogenesis stages of elongation, individualization, and coiling; these testis show aberrant DNA location, lack of actin structures, and increased apoptosis. Brca2KO heterozygous males do not show these defects.

Mutant flies are sensitive to camptothecin, topotecan and to ionising radiation.

Brca2KO/Brca256E larvae are highly sensitive to treatment with X-rays or with methyl methanesulfonate.

Brca2KO/Brca256E mutants show significant decreases in HR-h (homologous recombination using the homologous chromosome) repair in a "Repair Reporter 3" assay when the HR-h pathway is available. Short tract HR-h is decreased 64-fold from wild-type values and long-tract HR-h in decreased 9-fold. There is a significant increase in single-strand annealing and in flanking deletions in the mutant flies compared to controls under these conditions.

Eggs laid by Brca2KO/Brca256E females have a highly penetrant karyosome defect. Some of these eggs are weakly ventralised, having two dorsal appendages that are fused at the base.

Some eggs produced by homozygous females are weakly ventralised, either having two dorsal appendages that are fused at the base or having a single dorsal appendage.
