Description

Imprecise excision of the progenitor insertion has resulted in a 2925bp deletion that extends from +308 to +3232 relative to the ebd1 transcription start site. Approximately 3kb of the original insertion remains.

Comment:

2925 bp deletion resulting from the imprecise excision of P{EPgy2}ebd1EY01876. Extends from +308 to +3232 relative to the ebd1 transcription start site. 3L:685545 (R5) was used as the transcription start site for the mapping. Approximately 3kb of the original insertion remains.

The adult midgut defects (excess of mitotic cells, aberrant epithelium architecture) characteristic for Apc[Q8] mutants, a fly model of colorectal cancer, can be ameliorated by combination with ebd1[240]/Df(3L)ED9698, ebd2[136] mutant alleles.

Detailed Description

Adult ebd1240 mutants show significant increase in the number of progenitor cells (intestinal stem cells and enteroblasts) in the midgut and unlike in controls frequently display chains and less frequently even clusters of progenitor cells. ebd1240 mutant midgut somatic clones induced during adulthood are markedly larger and contain more cells than control clones and an abnormally large number of progenitor cells (due to excessive mitotic activity of the cells) is found in the vicinity of the mutant clones.

Mutant flies have defects in the dorsal longitudinal muscles of the thorax.

All mutant females and a third of mutant males lack one or more dorsal longitudinal muscles in the thorax, with the remaining dorsal longitudinal muscles being increased in size.

ebd1240 mutants show a reduction in the number of a subset of dorsal longitudinal muscles, and enlargement of the remaining indirect flight muscles.

Adult ebd1240/ebd1Df5,ebd2136 mutants show significant increase in the number of progenitor cells (intestinal stem cells and enteroblasts) in the midgut and unlike in controls frequently display chains or clusters of progenitor cells, the penetrance of this latter phenotype is increased further in ebd1Df5,ebd2136/ebd1240,ebd2136 double mutants.

The adult midgut epithelium defects (increased number of progenitor cells, cell polarity and adhesion defects along with epithelium multi-layering) can be rescued by combination with ebd1240/Df(3L)ED9698,ebd2136. The increased number of enteroblast cells in midguts of newly eclosed ApcQ8 mutant adults is significantly suppressed by ebd1240 homozygosity.

ebd1240/ebd1Df5 suppresses the apoptosis of photoreceptor cells which is seen in ApcQ8 homozygotes, such that the rescued photoreceptors are wild type in length.

ebd1240/Df(3L)ED4136 suppresses the shortened photoreceptor phenotype caused by expression of armS10.Scer\UAS.T:Hsap\MYC under the control of Scer\GAL4GMR.long.

One copy of ewg2 enhances the indirect flight muscle loss seen in ebd1240 mutant flies.

Expression of Hsap\JRKScer\UAS.cBa under the control of Scer\GAL4ebd1.PB rescues the indirect flight muscle defects seen in ebd1240 ebd2136/Df(3L)ED9698 ebd2136 mutants.
