Excision of the P{EP} element in P{EP}MAPk-Ak2GE3296 results in deletion of the region downstream of the insertion site, which includes the second exon and most of the third exon of MAPk-Ak2.
No obvious phenotypes are seen in MAPk-Ak2Δ43 homozygous mutants under normal conditions.
Approximately 38% of MAPk-Ak2Δ43 mutant larvae exhibit a melanotic phenotype in response to strong salt stress (0.2M NaCl). A "black dot" (BD) is seen in the posterior hindgut and the affected hindgut epithelium appears pathologically altered. Approximately 20% of mutants exhibit this phenotype when raised on food containing 0.1M NaCl and no phenotypes are observed when MAPk-Ak2Δ43 mutants are raised on normal food. The dorsal epithelial cells (ECs) containing BDs are apically ruptured, whereas no damage is detected in wild type hindguts under the same conditions. No morphological alterations are seen in MAPk-Ak2Δ43 mutant ECs that do not contain a BD, although a blistering of the surface without damage to the apical membrane is occasionally observed. TUNEL staining reveals that local clusters of apoptotic hindgut ECs are present in mutant larvae under salt stress conditions, and these are not present in controls. The visceral muscles surrounding the hindgut ECs appear unaffected, with a disturbed hindgut musculature only observed in hindguts with large BDs. Loss of epithelial integrity close to BDs is also observed, as determined by more diffuse Avic\GFPNrg-G00305 expression. BDs are only seen in the dorsal hindgut hd compartment.

MAPk-Ak2Δ43 Mpk21 double mutant larvae exhibit a weak melanotic phenotype under normal food conditions. A "black dot" is seen in the posterior hindgut in approximately 6% of larvae. The number of larvae displaying black dots increases in response to salt stress. The semi-lethality seen in Mpk21 mutants in response to salt stress is enhanced by MAPk-Ak2Δ43.
MAPk-Ak2Δ43 slightly enhances the melanotic phenotype seen in p38bd27 larvae under normal food conditions. A "black dot" (BD) is seen in the posterior hindgut in approximately 28% of larvae.
