UASt regulatory sequences drive expression of a 500bp inverted repeat.
Expression of Tango1NIG.11098R under the control of Scer\GAL4bs.Term causes air-filling defects and reduced branch numbers in the trachea terminal cells region. Terminal cells show abnormal ER and Golgi, as compared to controls: abnormal/enlarged ER exit sites (identified by Sec16 staining), which seem to be enclosed by the medial Golgi (identified by ManII-GFP) instead of being juxtaposed; the cis-Golgi (identified by GM130) and trans-Golgi (identified by GalT) shows an abnormal enlarged ring-like structure.
Expression under the control of Scer\GAL4bs.Term, Scer\GAL4nub.PK, Scer\GAL4repo.PU or Scer\GAL4sr-md710 impairs the secretion of Dpy by tracheal terminal cells, wing disc pouch cells, larval brain glial cells or pupal tendon cells, respectively; expression under the control of Scer\GAL4repo.PU also leads to the abnormal accumulation of LanA and LanB1 in the cytoplasm of glial cells; expressing under the control of Scer\GAL4nub.PK or Scer\GAL4ptc.PU induces a decrease in endoplasmic reticulum exit sites (identified by Sec16) in the wing disc pouch. Expression under the control of Scer\GAL4apolpp.PB, but not Scer\GAL4bs.Term, lead to a decrease in the levels of Collagen-IV surrounding tracheal terminal cells; expression under the control of Scer\GAL4apolpp.PB leads to accumulation of Collagen-IV, LanA, LanB1 and SPARC, as well as to endoplasmic reticulum exit site aggregates (identified by Sec16) in the larval fat body; both of these results indicate a defect in secretion by the fat body.
