Overexpression of DetS125E.Scer\UAS.P\T.T:Avic\GFP under the control of Scer\GAL4nos.UTR.T:Hsim\VP16 results in a significant increase in both the number of germline cells per cyst and the penetrance of the phenotype. Almost 50% of mutant egg chambers exhibit 32 cells or more, with oocytes displaying up to 6 ring canals.
Expression of DetS125E.Scer\UAS.P\T.T:Avic\GFP in dividing germline cysts, using the Scer\GAL4bam.T:Hsim\VP16 driver, which is expressed from cystoblasts to eight cell cysts but not in germline stem cells, does not result in any extra germline cells.
Live-imaging of germline stem cells expressing DetS125E.Scer\UAS.P\T.T:Avic\GFP under the control of Scer\GAL4nos.UTR.T:Hsim\VP16 reveals the germline stem cells to be dividing synchronously with several neighboring germline cells, suggesting that they may be physically connecting, which is never seen in a wild-type situation. The spectrosomes in these germline stem cells are branched as fusomes and linked germline stem cells with up to eight other germline cells. These synchronous cells appear to have characteristics of both stem cells and cyst cells. They do not grow indefinitely, and the fusome eventually breaks down, indicating that DetS125E.Scer\UAS.P\T.T:Avic\GFP delays abscission, but does not block it completely.

Reducing the level of CycB, through a CycB2/+ background, greatly enhances the Scer\GAL4nos.UTR.T:Hsim\VP16>DetS125E.Scer\UAS.P\T.T:Avic\GFP gain-of-function phenotype. Further reduction in the level of CycB, through a CycB2/CycBKG08886 background, greatly enhances the Scer\GAL4nos.UTR.T:Hsim\VP16>DetS125E.Scer\UAS.P\T.T:Avic\GFP gain-of-function phenotype.
