4bp deletion (nucleotides 186-189) and 12bp deletion (nucleotides 419-431), both within Vps8. The 4bp deletion results in a frameshift and a premature stop codon which is predicted to result in the translation of only the first 39 amino acid residues of Vps8.

Vps81/Df(3L)ED211 transheterozygous garland cells are enlarged and show smaller late endosomes (Rab7-positive puncta) than controls, but do not show an obvious increase in the number of autophagosomes (endogenous Atg8a-positive puncta).

Vps81/Vps81 and Vps81/Df(3L)ED211 mutant flies are semilethal, with most dying as pharate adults. Dissected nephrocytes of Vps81/Df(3L)ED211 and Vps81/Vps81 larvae display defective endosomal trafficking as shown by incubation with, and subsequent analysis of internalization and transport of fluorescent dextran. Garland nephrocytes in these mutants also exhibit late endosome and endolysosome fragmentation, and absent α-vacuoles, but early endosomes are unaffected.

Surviving Vps81/Df(3L)ED211 adults are weak, fail to unfold their wings properly, and die within 24 hr, but do not display any eye pigmentation defects. Vps81/Df(3L)ED211 mutant larvae have significantly more circulating hematocytes than controls, and often contain hemocyte-derived melanotic tumors in their body cavity, disorganization of the sessile hemocyte compartment, and accumulation of crystal cells and lamellocytes. Hemocytes of these mutants fail to acidify bacteria-containing phagosomes upon injection of E. coli. In an assay of nephrocyte function using food supplementation with silver nitrate, Vps81/Df(3L)ED211 larvae exhibit impaired silver uptake and endo-lysosomal trafficking. The endosomal compartment of Vps81/Df(3L)ED211 mutant wing imaginal discs is indistinguishable from wild type.

ltLL07138/Df(2L)lt45, Vps81/Df(3L)ED211 double mutants exhibit similar melanotic tumor-formation and nephrocyte late endosome fragmentation phenotypes as compared to Vps81/Df(3L)ED211 single mutants, and nephrocytes also exhibit many small electron dense lysosome-like organelles.
