THADAKO1 mutants are viable and have normal developmental patterning, timing, adult wings size, wing cell size and lifespan compared to controls but display metabolic defects, are hyperphagic and obese (measured as a fat/protein ratio), although the robustness of the increased-feeding phenotype varies considerably with genetic background; the lipid droplets in the fat body cells of THADAKO1 adults are larger and take up larger proportion of the cell area compared to controls, the flies however do not display any significant ectopic lipid deposition in guts. The adult flies survive significantly longer upon starvation compared to controls but show impaired recovery from cold-induced paralysis and generate less heat per unit of body biomass.
The obesity (measured as fat/protein ratio) of THADAKO1 mutant adults is rescued by expression of SERCAKK107371 driven by Scer\GAL4da.PU and partially so by Scer\GAL4tub.PU-controlled expression of Itp-r83AScer\UAS.cVa.
The obesity (measured as fat/protein ratio) of THADAKO1 mutant adults is rescued by expression of Hsap\THADAScer\UAS.cMa driven by Scer\GAL4hs.PU.
THADAKO1 is partially rescued by THADAUAS.cMa/Scer\GAL4elav.PU
THADAKO1 is partially rescued by THADAUAS.cMa/Scer\GAL4da.PU
THADAKO1 is partially rescued by THADAUAS.cMa/Scer\GAL4mir-278-Gal4KI
THADAKO1 is partially rescued by THADAUAS.cMa/Scer\GAL4Adh.PF
The obesity (measured as a fat/protein ratio) as well increased food intake characteristic for THADAKO1 mutant adults are partially rescued by expression of THADAScer\UAS.cMa controlled by either Scer\GAL4elav.PU or Scer\GAL4da.PU. The increased adiposity of THADAKO1 mutants is also improved by Scer\GAL4mir-278-Gal4KI (while the increased food intake is not rescued) or Scer\GAL4Adh.PF-driven expression.
The poor recovery from cold-induced paralysis displayed by THADAKO1 mutants is rescued by expression of THADAScer\UAS.cMa under the control of Scer\GAL4da.PU in the mutant background.
