Detailed Description

Whole eye ifcKO homozygous clones in an otherwise mosaic individual show a progressive loss of rhabdomeres accompanied with distorted photoreceptor margins when under constant light conditions. Likewise, electroretinogram depolarization readings progressively decrease when under constant light conditions. These defects are not observed at eclosion nor under constant darkness. Clones within mosaic eyes also show the same significant decrease in rhabdomere number, associated with apoptotic bodies and increased lipid droplet-containing autophagosomes (autophagosomes: Atg8a-positive puncta; lipid-droplets: Bodipy-positive puncta).

Whole-eye ifcKO homozygous clones also show a significant increase in the number of lipid droplets (Bodipy-positive puncta), as compared to controls.

Expressing ifcUAS.cJa under the control of Scer\GAL4αTub84B.PL partially rescues the second instar larval lethality of ifcKO homozygotes, so that most individuals reach the pupal stage or adulthood; expressing under the control of either Scer\GAL4elav.PLu, Scer\GAL4repo or Scer\GAL4Mef2.PR only allows some individuals to reach pupal stage or adulthood.

Expressing ifcUAS.mCherry under the control of Scer\GAL4αTub84B.PL partially rescues the second instar larval lethality of ifcKO homozygotes, so that most individuals reach the pupal stage or adulthood; expressing under the control of either Scer\GAL4elav.PLu, Scer\GAL4repo or Scer\GAL4Mef2.PR only allows some individuals to reach pupal stage or adulthood.

Notes on Origin
Discoverer

Separable from second-site mutation(s) on the chromosome, affecting the MED15, lwr, Nle and Sf3b1 complementation groups. Since these four genes reside within approximately 90kb near the telomere in 2L, the second site lesion may be a small deletion that removes all four genes.
