CycEAR95 produces tiny or no transient follicle cell clones 5 days after clone induction and no follicle stem cell clones 12 days after clone induction.

In CycEAR95 mutants, both NB6-4t daughter cells differentiate as glial cells as opposed to one glial cell and one neuronal precursor as in wild-type. The NB6-4t lineage is transformed to NB6-4a in approximately 65% of thoracic hemisegments. The NB6-4a lineage is not affected in CycEAR95 mutants.

37.7% of egg chambers in CycEJP/CycEAR95 flies raised at 18oC, have half the normal number of germ-line cells.

CycEAR95/CycEJP animals have small, rough eyes.

CycEAR95 mutant embryos show a size reduction of the large intestine region.

CycEJP/CycEAR95 flies have more severe eye defects than CycEJP homozygotes.

S phase is widely induced in the internal tissues that endocycle in wild-type embryos in CycEAR95 embryos carrying two copies of CycAhs.PK, within 1 hour of heat shock. Induction of S phase is also seen in epidermal cells, although not until 180 minutes after heat shock.

Heterozygotes exhibit normal eyes.

Homozygotes exhibit wild type eyes.

After embryonic mitosis 16, cell cycle progression is clearly arrested. BrdU incorporation experiments show that endoreplicative as well as mitotic S phases are arrested.

wor4/+, CycEAR95/+, Df(3R)Exel6186/+, Df(3R)Exel6212/+ quadruple heterozygous stage 17 embryos show defects in neuroblast in lineage progression, as illustrated by the frequent loss of the last-born cell in the NB5-6T lineage, as compared to controls.

The wing nicking phenotype caused by expression of Rbf280.Scer\UAS under the control of Scer\GAL4bbg-C96 is enhanced if the flies are also carrying CycEAR95.

Heterozygosity for either CycE5 or CycEAR95 suppresses the small bristle phenotype of mi1/miΔEY22 flies.

Scer\GAL4tub- and Scer\GAL4Act5C.PI-driven co-expression of E2fScer\UAS.cNa and DpScer\UAS.cDa in CycEAR95 follicle stem cell (FSC) clones also expressing CycEWX.Scer\UAS.cWa suppresses the CycEAR95-associated defective FSC-maintenance phenotype.

The reduced size of the posterior compartment of the wing that is seen in animals expressing crolScer\UAS.cMa under the control of Scer\GAL4en-e16E is suppressed if they are also heterozygous for CycEAR95.

A heterozygous CycEAR95 background strongly enhances the dorsalisation phenotype seen when ImpScer\UAS.cGa is expressed under the control of Scer\GAL4mat.αTub67C.T:Hsim\VP16 (on the second).

In CycAC8LR1; CycEAR95 double mutant embryos, cells arrest before mitosis 15, a phenotype not seen in either single mutant.

The homeotic transformation of NB6-4a to NB6-4t, observed in abd-AMX1 mutants is suppressed in CycEAR95 mutants.

Adult viability of CycD1/Y ; CycEAR95/+ males is only 35% of expected value. Adult viability of CycD2/Y ; CycEAR95/+ males is only 18% of expected value.

The number of egg chambers with twice the normal number of germ cells in CSN5ex21/CSN5L4032 flies, is reduced to 0.6% by CycEAR95/+ (n>500).

The addition of CycEAR95 to E2f276Q1/E2f2p111-5, E2fsu89, partially suppresses the salivary gland endoreplication defect. Salivary gland nuclei in these animals have about 70% of the DNA content of wild-type. The lethality is also partially suppressed - 52% of animals survive to the pupal stage (compared to 25%). Finally the macrochaetae phenotype is strongly suppressed. - 99% of flies show normal macrochaetae (as opposed to 12%).

The small, rough eye phenotype of CycEAR95/CycEJP animals, is almost completely dominantly suppressed by hpo42-47.

The expression of ruxhs.PT during G2-phase 14 in CycEAR95 mutants leads to a degradation of Cyclins, apparently resetting G2 cells to G1. Nuclei retain interphase 14 size and density and exhibit increased ploidy. Tetranemic chromosomes are also generated.

Enhances the mild rough eye phenotype of dapGMR.PdN flies. The dapGMR.PdN CycEAR95 phenotype is suppressed by spenpoc361 and eye morphology is restored.

Mitosis is inhibited when rapScer\UAS.cSa is expressed in CycEAR95 embryos using Scer\GAL4prd.RG1.

CycEAR95 heterozygotes carrying four copies of RbfGMR.PD exhibit eyes with fused ommatidia and missing bristles.

In a CycEAR95 mutant background, no DNA replication is induced in stage 13 or 14 embryos by simultaneous over-expression of E2fhs.PD and Dphs.PD. In a heterozygous CycEAR95 mutant background simultaneous over-expression of E2fhs.PD and Dphs.PD causes induction of S phase in the salivary gland and midgut, but not the epidermis.

Expression of dapGMR.PdN transgene in CycEAR95 flies causes a rough eye phenotype.

Xenogenetic Interactions
Statement
Reference

CycEAR95 enhances the eye phenotypes seen when Mmus\Shisa5Scer\UAS.cGa is expressed under the control of Scer\GAL4dpp.PH.

The eye phenotype of flies carrying a weakly expressing P{GMR-BZLF1.A} line is unaffected by CycEAR95.
