Pan-neural expression of NetBScer\UAS.cHa under the control of Scer\GAL4sca-109-68 dramatically reduces apoptosis in the thoracic and abdominal segments of the ventral nerve cord.

When Df(1)NetABΔ mutant females expressing NetBScer\UAS.cHa under the control of Scer\GAL4sli.PS are crossed to wild type males there is a reduction in the proportion of eggs that hatch compared to wild type.

Expression of NetBScer\UAS.cHa using Scer\GAL4sli.PS results in the dendrites of the motoneuron labelled by Scer\GAL4VGlut-VGN9281 having a wild type distribution.

The projections from the pCC neurons on either side of the midline are attracted toward the midline in 11 hour embryos expressing NetBScer\UAS.cHa under the control of Scer\GAL4unspecified. 14 hour embryos expressing NetBScer\UAS.cHa under the control of Scer\GAL4unspecified show collapse of the longitudinal tracts at the midline in 40% of hemisegments, and in other hemisegments the longitudinal tracts are shifted closer to the midline.

When expression is driven by Scer\GAL4GMR.PF, retinal axons stall in the optic stalk, spread abnormally or criss-cross each other along the D/V axis. Photoreceptor axons also collapse onto each other along the A/P axis. They fail to innervate their target region properly. When expression is driven in clones in the anterior, undifferentiated portion of the eye disc or in the lamina anlage by Scer\GAL4Act5C.PP there is no effect on retinal fibers. However when clones fall in the differentiating portion of the lamina, mild hypoinnervation and irregular spacing of retinal fibers occurs.

Scer\GAL4F63-mediated expression elicits inappropriate innervation by the transverse nerve (ectopic innervation of muscles 7 and 6 and occasionally 13). Scer\GAL4how-24B-mediated expression sometimes causes the entire SN to stall as a clump of growth cones either within the CNS or in the ventral muscle region The SNc often extends for only a short distance and then ends in a clump or fails altogether. When the SN reaches the lateral muscle domain the SNa often fails to branch. Loss of Sema-2a expression allows both the SNa and SNc to project normally. Scer\GAL4A51-mediated expression in muscle 5 specifically repels the lateral branch of the SNa, the nerve sometimes takes a circuitous path to muscle 8 without innervating muscle 5. Scer\GAL4how-24B-mediated expression allows the axons of the transverse nerve to ectopically innervate muscles 7 and 6.

Df(1)KA9 and Df(1)RK5 embryos exhibit variable defects in the formation of commissural pathways, Scer\GAL4rho.PL induced expression of P{UAS-NETB} significantly rescued the CNS phenotype. Scer\GAL4sca-537.4 induced expression leads to a variety of CNS defects; reductions in the number of axons in commissural bundles and more extremely missing commissures.

The longitudinal tracts of embryos expressing NetBScer\UAS.cHa under the control of Scer\GAL4unspecified in a sliE-158 background appear similar to those seen in strong loss-of-function sli mutant embryos.

NetBScer\UAS.cHa; Scer\GAL4sim.PS largely suppresses the reduction of ventral midline commissures in sizU112 embryos.

Expression of NetBScer\UAS.cHa under the control of Scer\GAL4605 in fra3/fra4 embryos results in abnormal spreading of the dMP2 growth cone over the region of ectopic NetB expression.

Expression of NetBScer\UAS.cHa under the control of Scer\GAL4sim.P3.7 completely rescues the axon scaffold defects seen in stage 16 Df(1)NetABΔ mutant embryos, although a few minor defects are still visible. Expression of NetBScer\UAS.cHa under the control of Scer\GAL4rho.NEE does not rescue the phenotypes, and commissural and longitudinal defects are still present. Expression of NetBScer\UAS.cHa in post-mitotic neurons under the control of Scer\GAL4AD.elav.T:Zzzz\ZipEERRL,T:SV40\nls2 also produces a high degree of rescue. The increased apoptosis seen from stage 14 onwards is also suppressed when NetBScer\UAS.cHa is expressed under the control of Scer\GAL4AD.elav.T:Zzzz\ZipEERRL,T:SV40\nls2.

Expression of NetBScer\UAS.cHa under the control of Scer\GAL4rho.NEE rescues the negative geotaxis defects seen in Df(1)NetABΔ mutant flies. Some rescue is seen when NetBScer\UAS.cHa is expressed under the control of Scer\GAL4sim.P3.7, but no rescue is seen with the Scer\GAL4AD.elav.T:Zzzz\ZipEERRL,T:SV40\nls2 driver. Significant rescue is also seen when NetBScer\UAS.cHa is expressed without a Scer\GAL4 driver, but it is suggested that this result is due to a single exceptional fly. The mechanical-startle induced locomotor defects seen in Df(1)NetABΔ cannot be suppressed under the control of any of Scer\GAL4AD.elav.T:Zzzz\ZipEERRL,T:SV40\nls2, Scer\GAL4sim.P3.7 or Scer\GAL4rho.NEE, although expression of the transgene was able to increase the locomotor phenotype even further.

Expression of NetBScer\UAS.cHa under the control of Scer\GAL4sim.P3.7 rescues the semi-viability associated with Df(1)NetABΔ. The fertility defects are not rescued under the control of any of Scer\GAL4AD.elav.T:Zzzz\ZipEERRL,T:SV40\nls2, Scer\GAL4Ilp7.PY, Scer\GAL4rho.NEE, Scer\GAL4sca-109-68, Scer\GAL4sim.P3.7, Scer\GAL4sli.PS or Scer\GAL4Tdc2.PC.

Expression of NetBScer\UAS.cHa under the control of any of Scer\GAL4AD.elav.T:Zzzz\ZipEERRL,T:SV40\nls2, Scer\GAL4rho.NEE, Scer\GAL4sca-109-68 or Scer\GAL4sim.P3.7 has no effect on the male courtship 'mirror movement' phenotype seen in Df(1)NetABΔ mutants.

Expression of NetBScer\UAS.cHa under the control of either Scer\GAL4AD.elav.T:Zzzz\ZipEERRL,T:SV40\nls2 or Scer\GAL4sca-109-68 is unable to rescue the flightlessness seen in Df(1)NetABΔ mutant flies.
