Detailed Description

barrL305 homozygotes and barrE831/barrL305 transheterozygotes show variable embryonic muscle pattern defects, which may include: misshaped and disoriented muscles, missing muscles and/or "holes" in the muscle pattern; distorted pharynx musculature; heart cells are occasionally missing or duplicated. These musculature defects are also observed in barrL305/Df(2L)Exel7077 transheterozygotes that also express barrUASp.cOa.EGFP under the control of either Scer\GAL4sns.PK or Scer\GAL4kirre-rP298-G4, but not when under the control of either Scer\GAL4twi.PB or Scer\GAL4Mef2.PR.

Average Citrate Synthase activity is increased by 4% in heterozygous males

Homozygous embryos exhibit severe loss of PNS neurons and a significant increase in size of remaining neurons. Embryos arrest in cycle 16, in the majority of anaphase figures chromatids do not resolve but remain interlocked to form extensive chromatin bridges. The unresolved chromatids decondense without segregation and cytokinesis is attempted. Trl staining reveals mutation does not prevent centromere separation.

barrL305, Df(3L)BSC395 double homozygous embryos show disorganized somatic muscles, which are drastically reduced in mass and number, as compared to controls.

Intermediate eye pigment levels are observed in glu88-82, barrL305 double mutants with a In(1)wm4h background, showing that glu88-82 suppressor effects of PEV are not dominant over the enhancer activity of barrL305.

Chromatin condenses nonuniformly in prophase and prometaphase in postblastoderm Cap-GK1 barrL305 double mutant embryos. By metaphase, chromosome condensation has reached wild-type levels. Chromatid separation defects are seen in anaphase when only the fourth chromosome is clearly seen to separate, and lagging chromosomes are seen in telophase. The double mutant postblastoderm embryos have an increased number of prometaphase figures compared to wild-type embryos, suggesting that the length of prometaphase may be increased in the mutant embryos.
