Expression of IswiK159R.Scer\UAS.T:Ivir\HA1 under the control of Scer\GAL4ey.PH results in a rough eye phenotype.

Expression of IswiK159R.Scer\UAS.T:Ivir\HA1 under the control of Scer\GAL4ey.PH results in global decondensation of the polytene chromosomes.

Expression of IswiK159R.Scer\UAS.T:Ivir\HA1 under the control of Scer\GAL4ey.PH does not cause obvious changes in chromosome size. The DNA content of the chromosomes is reduced in the mutants compared to wild type. When normalised for DNA content, the volume of the polytene chromosomes of the mutant larvae is indistinguishable from the volume of polytene chromosomes of control larvae. The banding pattern is often disrupted, and "holes" which may represent regions of decondensed chromatin are seen in the mutant chromosomes.

Expression of IswiK159R.Scer\UAS.T:Ivir\HA1 under the control of Scer\GAL4ey.PH in an otherwise wild-type background results in flies with rough and reduced eyes. The severity of the phenotype is increased when IswiK159R.Scer\UAS.T:Ivir\HA1 is expressed under the control of Scer\GAL4ey.PH in an Iswi2 background.

Expression of IswiK159R.Scer\UAS.T:Ivir\HA1 under the control of Scer\GAL4ey.PH results in a dramatic decondensation of the X chromosome and the autosomes in polytene chromosome preparations from larvae of both sexes.

Expression of IswiK159R.Scer\UAS.T:Ivir\HA1 under the control of Scer\GAL4da.G32 dramatically alters the structure of metaphase chromosomes prepared from neuroblasts of third instar larvae. The chromatin appears highly decondensed and disorganised and sister chromatids are often indistinguishable from each other. The euchromatin appears to be most strongly affected, appearing hazy and diffuse, while the largely heterochromatic fourth and Y chromosomes show relatively normal condensation.

Animals expressing IswiK159R.Scer\UAS.T:Ivir\HA1 under the control of Scer\GAL4da.G32 and derived from Iswi2/+ females survive until late larval stages at 18[o]C but do not complete embryogenesis at 25[o]C. At 25[o]C, the embryos show defects in chromosome condensation and in the organisation of the mitotic spindle as early as nuclear cycle 12.

Overexpression of IswiK159R.Scer\UAS.T:Ivir\HA1 under the control of Scer\GAL4ap-md544 leads to a loss of sensory organs, including a frequent loss of dorsocentral bristles. Overexpression of IswiK159R.Scer\UAS.T:Ivir\HA1 under the control of Scer\GAL4pnr-MD237 reduces the numer of dorsocentral bristles (an average of 1.56 bristles per heminotum compared to 2.0 in wild-type).
