hfpEP3058 homozygous mutant larvae and pupae are significantly larger than wild-type controls and display an increase in the number mitotic cells and cells in the S-phase of the cell cycle in the third instar larval wing disc.

Pupariation in pUf68EP3058 homozygous animals is delayed by approximately two days. The resulting wandering third instar larvae and pupae are about 20% larger than wild-type controls. The duration of pupariation is normal but these animals fail to eclose and die as pharate adults that are larger than wild-type, have duplication of the superior scutellar macrochaete and malformation of legs, wings and sex combs. The number of S phase cells and mitotic cells is increased in pUf68EP3058 mutant wing discs: BrdU incorporation is uniform across the disc and cell cycle arrest is not evident in the zone of non-proliferating cells found at the dorsal-ventral compartment boundary of wild-type late third instar wing-discs. In pUf68EP3058/pUf68EP3058 eye discs the S phase band posterior to the morphogenetic furrow is broader than in wild type, but the second mitotic wave does not occur prematurely. Despite the increased proliferation in these discs, they are not overgrown and there is no obvious difference in cell size compared to wild-type. There are, however, around twice as many apoptotic cells in the wing imaginal discs of these mutants than in wild-type. Dividing cells in Cycle 16 pUf68EP3058/pUf68EP3058 embryos exhibit abnormal chromosome morphology, including many lagging chromosomes that are often mis-segregated due to closure of the contractile ring prior to sister chromatid separation.

pUf68EP3058/Df(3L)Ar14-8 animals are slower to develop than controls and die shortly after pupariation with melanised patches of tissue.

Unlike stgAR2 homozygotes, stgAR2; pUf68EP3058 double homozygotes survive embryogenesis and die between first and second instar.
