Pupae expressing Prosβ61.B.UAS under the control of Scer\GAL4how-24B raised at the non-permissive 25[o]C temperature lack the middle thoracic portion of their bodies, indicating degeneration of muscle tissue. There are no obvious defects at 18[o]C.

Expression of Prosβ61.B.Scer\UAS under the control of Scer\GAL4GMR.long does not induce rough eye phenotype in adult flies maintained at 28[o]C.

Expression of temperature-sensitive Prosβ61.B.Scer\UAS under the control of Scer\GAL4GMR.PF results in rough eye phenotype at the non-permissive temperature of 28[o]C, at the permissive temperature (22[o]C) the eyes appear normal.

Co-expression of Prosβ21.Scer\UAS and Prosβ61.B.Scer\UAS under the control of Scer\GAL4wor.PA does not induce larval neuroblast loss.

Expression of Pros261.B.Scer\UAS at 29oC under the control of Scer\GAL4GMR.PF results in severely disrupted eyes, with fused facets and small spots of black, presumably necrotic, cells.

Flies co-expressing Prosβ61.B.Scer\UAS and Prosβ21.Scer\UAS under the control of Scer\GAL4elav.PU at the non-permissive temperature 30[o]C are lethal at the third instar larval stage. When the flies are raised at 18[o]C and then incubated at 30[o]C for 4 weeks and exposed to the pesticides paraquat and/or maneb no dopaminergic neuron loss is observed.

Co-expression of Prosβ61.B.Scer\UAS and Prosβ21.Scer\UAS in the salivary gland under the control of Scer\GAL4fkh.PH causes aberrant morphology of some salivary gland nuclei.

Co-expression of Prosβ61.B.Scer\UAS and Prosβ21.Scer\UAS under the control of Scer\GAL4GMR.PU at 29[o]C results in a rough eye phenotype. This phenotype is enhanced if the animals are also co-expressing either TnksGD11407 or CG9588HM05013.

However, the pseudopupil images of flies expressing Pros261.B.Scer\UAS and Prosβ21.Scer\UAS reveal severe retinal damage, which is substantially rescued by additional co-expression of one copy of P{Sym-UAS-Hsrω}.

Expression of Pros261.B.Scer\UAS and Prosβ21.Scer\UAS in the developing tracheal system under the control of Scer\GAL4btl.PS is sufficient to cause dorsal trunk breaks due to down-regulation of trh in embryonic fusion cells.

The eye degeneration phenotype observed in adult flies expressing either Zzzz\CGG90.Scer\UAS.T:Avic\GFP-EGFP or Zzzz\CGG100.5'UTR.Scer\UAS.T:Avic\GFP under the control of Scer\GAL4GMR.long (and maintained at 28[o]C) is significantly enhanced by co-expression of Prosβ61.B.Scer\UAS.

Co-expression of the temperature sensitive Prosβ61.B.Scer\UAS allele together with Hsap\MAPTScer\UAS.T:Avic\GFP under the control of Scer\GAL4shot-OK307 has no adverse effect on the morphology and structure of the adult thorax or the thoracic somatic muscles below at either the non-permissive (28[o]C) or the permissive (22[o]C) temperatures.

Simultaneous co-expression of Prosβ61.B.Scer\UAS and Prosβ21.Scer\UAS does not suppress the protective role of expression of Hsap\ATXN3Scer\UAS.WT.T:Zzzz\His6 or DnaJ-1Scer\UAS.cKa in suppression of eye degeneration in Scer\GAL4GMR.long>Hsap\ATXN3tr.Q78.Scer\UAS.T:Ivir\HA1 flies.

Simultaneous co-expression of Prosβ61.B.Scer\UAS and Prosβ21.Scer\UAS leads to lethality in Scer\GAL4GMR.long>Hsap\ATXN3tr.Q78.Scer\UAS.T:Ivir\HA1 flies; extra co-expression of Hsap\ATXN3Scer\UAS.WT.T:Zzzz\His6 or DnaJ-1Scer\UAS.cKa suppresses this lethality.

The eye damage caused by Scer\GAL4GMR.PF-mediated expression of Zzzz\CAG127Q.Scer\UAS.T:Ivir\HA1 is enhanced by co-expression of Pros261.B.Scer\UAS and Prosβ21.Scer\UAS. Co-expression of P{Sym-UAS-Hsrω} still suppresses the damage in Scer\GAL4GMR.PF, Zzzz\CAG127Q.Scer\UAS.T:Ivir\HA1, Pros261.B.Scer\UAS, Prosβ21.Scer\UAS eyes, but the eyes are not rescued to the degree seen in Scer\GAL4GMR.PF, Zzzz\CAG127Q.Scer\UAS.T:Ivir\HA1 eyes.

The increase in inclusion bodies in larval eye discs caused by Scer\GAL4GMR.PF-mediated expression of Zzzz\CAG127Q.Scer\UAS.T:Ivir\HA1 is further increased by co-expression of Pros261.B.Scer\UAS and Prosβ21.Scer\UAS. However, co-expression of P{Sym-UAS-Hsrω} fails to reduce the number of inclusion bodies in the Scer\GAL4GMR.PF, Zzzz\CAG127Q.Scer\UAS.T:Ivir\HA1, Pros261.B.Scer\UAS, Prosβ21.Scer\UAS background.

The damaged eye phenotype that results from Scer\GAL4GMR.PF-mediated expression of Hsap\MJDtr.Q78.Scer\UAS.T:Ivir\HA1 is not further enhanced by co-expression of Pros261.B.Scer\UAS and Prosβ21.Scer\UAS. Co-expression of P{Sym-UAS-Hsrω} still suppresses the damage in Scer\GAL4GMR.PF, Hsap\MJDtr.Q78.Scer\UAS.T:Ivir\HA1, Pros261.B.Scer\UAS, Prosβ21.Scer\UAS eyes, but the eyes are not rescued to the degree seen in Scer\GAL4GMR.PF, Hsap\MJDtr.Q78.Scer\UAS.T:Ivir\HA1 eyes.

Co-expression of Prosβ61.B.Scer\UAS does not enhance the eye degeneration phenotype caused by expression of Hsap\ATXN3tr.Q78.Scer\UAS.T:Ivir\HA1 under the control of Scer\GAL4GMR.PU.

In the presence of Prosβ61.B.Scer\UAS, CG11700EP1384 cannot suppress the eye degeneration phenotype caused by expression of Hsap\ATXN3tr.Q78.Scer\UAS.T:Ivir\HA1 under the control of Scer\GAL4GMR.PU.
