Detailed Description

mir-9aE39/mir-9aJ22 mutant flies have a reduced body length, wing length and pupal volume. Haemolymph glucose and trehalose levels are also reduced.

Homozygotes and mir-9aJ22/mir-9aE39 animals lack approximately 50% of the posterior wing margin in all individuals and approximately 10% of animals also show mild loss of anterior wing margin.

Abundant cell death is seen in the wing pouch region of the wing disc in mir-9aJ22/mir-9aE39 third instar larvae, in contrast to wild-type controls. There is substantially more cell death in the dorsal compartment than in the ventral compartment. Cell death is not seen in the pronotum region of the disc in the mutant larvae.

mir-9aJ22/mir-9aE39 but not homozygous adults show a mild increase in notum microchaete density compared to controls.

mir-9aJ22/mir-9aE39 adults often have ectopic dorsocentral and anterior postalar bristles.

Wings carrying large homozygous clones show notching along the posterior wing margin.

Small homozygous clones in the wing disc contain apoptotic cells.

37% of mir-9aJ22 third instar larvae have an extra ddaE or ddaF neuron in one or two but not other hemisegments.

One copy of sNPF-RMI00427 slightly suppresses the reduction in body length seen in mir-9aE39/mir-9aJ22 mutant flies.

mir-9aE39/+ significantly enhances the supernumerary scutellar bristle phenotype of TBPHQ367X/+ flies.

Expression of thScer\UAS.cHa under the control of Scer\GAL4ap-md544 completely rescues the wing notching phenotype of mir-9aJ22/mir-9aE39 animals.

sensE2/+ results in an approximately 35% reduction in the loss of wing margin caused by mir-9aJ22/mir-9aE39. sensE2/+ does not rescue the increased cell death seen in the wing discs of mir-9aJ22/mir-9aE39 animals.

BxhdpR26/+ fully rescues the adult wing margin defects of mir-9aJ22 homozygotes and mir-9aJ22/mir-9aE39 animals. BxhdpR26/+ strongly suppresses the ectopic apoptosis seen in the wing discs of mir-9aJ22/mir-9aE39 animals.

The presence of BxhdpR26/+ partially suppresses the formation of ectopic anterior postalar bristles seen in mir-9aJ22/mir-9aE39 adults, but has only mild effects on the formation of ectopic dorsocentral bristles seen in these flies.

sensE2/+ partially suppresses the formation of ectopic dorsocentral and anterior postalar bristles seen in mir-9aJ22/mir-9aE39 adults.

Expression of BacA\p35Scer\UAS.cHa under the control of Scer\GAL4ap-md544 completely rescues the wing notching phenotype of mir-9aJ22/mir-9aE39 animals.

Comments

Expression of mir-9aScer\UAS.T:Disc\RFP-DsRed2.cSa under the control of Scer\GAL4Ilp2.PR rescues the reduction in body and wing length seen in mir-9aE39/mir-9aJ22 mutant flies.

Expression of mir-9aScer\UAS.cLa under the control of Scer\GAL4ap-md544 completely rescues the continuity of the wing margin in mir-9aJ22/mir-9aE39 animals. In the developing wing disc, the increased apoptosis seen in the wing pouch in mir-9aJ22/mir-9aE39 animals is rescued in the dorsal compartment by expression of mir-9aScer\UAS.cLa under the control of Scer\GAL4ap-md544.
