The expression of CkaGD12225 under the control of Scer\GAL4Mef2.PR leads to indirect flight muscle atrophy during pupal development (after 32h APF); fibers are longer than in controls and fail to compact at 32h APF.

Expressing CkaGD12225 under the control of Scer\GAL4C587 (together with Dicer-2, for a more efficient RNAi) leads to L3 male gonad defects: small gonads with supernumerary strongly DAPI-positive cells throughout, whereas control gonads exhibit brighter DAPI-stained DNA in the apical region of the tissue, where the GSCs, gonialblasts, TA spermatogonia, CySCs and CyCs reside. Further analysis uncovers additional defects in germline differentiation and somatic cell enclosure of cyst.

Adults expressing CkaGD12225 under the control of the cardioblast-specific Scer\GAL4tin.CΔ4 driver show significantly reduced survival on day 6 after a shift to 29[o]C compared to control flies.

Adults expressing CkaGD12225 under the control of Scer\GAL4elav.PLu (in the presence of Dcr-2Scer\UAS.cDa to increase the efficiency of RNAi) do not show a significant defect in avoidance of noxious temperature (46[o]C) compared to control flies.

Expression of CkaGD12225 under the control of Scer\GAL4elav.PLu (in the presence of Dcr-2Scer\UAS.cDa to increase the efficiency of RNAi) results in viable flies or partial lethality, depending on the particular P{GD12225} insertion line used.

Depending on the insertion line used, expression under the control of Scer\GAL4Mef2.PR can result in early pupal lethality or late pupal lethality.
