The expression of Ten-mGD3620 under the control of Scer\GAL4Or47b.7.467 has no significant effect on the pre-synaptic sites of adult olfactory receptor neurons Or47b, as assessed by the number and density of an active zone marker, as compared to controls.

The expression of Ten-mGD3620 under the control of Scer\GAL4Mz19 leads to the antennal lobe projection neurons DA1 lPN and VA1d adPN having significant decreases in the number and density of post-synaptic sites, but similar dendrite volume, as compared to controls; this expression also leads to the adult olfactory receptor neurons Or67d, which enervate the adult antennal lobe glomeruli DA1, having a significant decrease in the number of pre-synaptic sites, as assessed by an active zone marker, as compared to controls.

The expression of Ten-mGD3620 under the control of either Scer\GAL4221, Scer\GAL4arm.PU, or Scer\GAL4Ten-m-NP6658, in combination with Dicer-2 for efficient RNAi, results in the secondary dendrite branches of third instar larval class I abdominal dorsal multidendritic ddaE neurons being significantly less directionally biased towards the posterior, as compared to controls; expression under the control of Scer\GAL4221, in combination with Dicer-2 for efficient RNAi, also results in the secondary dendrite branches of third instar larval class I abdominal posterior ventral multidendritic vdap neurons being significantly less directionally biased towards the posterior, as compared to controls.

The expression of Ten-mGD3620 under the control of Scer\GAL4ppk.PG, in combination with Dicer-2 for efficient RNAi, results in third instar larval class IV abdominal dorsal multidendritic ddaC neurons showing no significant alteration in dendrite length, number of branch terminals, or spatial disposition of branches within the dendritic tree.

Expression of Ten-m[GD3620] under the control of either Scer\GAL4ppk.PG or Scer\GAL4Ten-m-NP6658, in combination with Dicer-2 for efficient RNAi, results in dendrite terminal branches of third instar larval class IV abdominal dorsal multidendritic ddaC neurons exhibiting a significantly less radial directional distribution, as compared to controls; expression under the control of Scer\GAL4hh-Gal4, in combination with Dicer-2 for efficient RNAi, also results in the dendrite terminal branches of third instar larval class IV abdominal dorsal multidendritic ddaC neurons exhibiting a significantly less radial directional distribution, but only in the dendritic tree region over an Hh-positive epidermal domain, as compared to controls.

Expression of Ten-mGD3620 in the olfactory receptor neurons (ORNs) under the control of Scer\GAL4peb-GAL4 results in mild intermingling of Ncra\QFMz19.PH-expressing projection neuron (PN) dendrites and Or47b-expressing ORN axons (these axons and dendrites do not overlap in wild type).

Expression of Ten-mGD3620 in the PNs (under the control of Scer\GAL4GH146) does not result in intermingling of Ncra\QFMz19.PH-expressing PN dendrites and Or47b-expressing ORN axons.

Expression of Ten-mGD3620 under the control of Scer\GAL4Mef2.247 results in a reduction in bouton number at the larval neuromuscular junction.

Expression of Ten-mGD3620 under the control of Scer\GAL4Ten-m-NP6658 results in the failure of muscle 3 innervation in 11% of hemisegments. This defect is still seen in the flies are also carrying either Scer\GAL80elav.PSb or Scer\GAL80Mhc.PW. The innervation of muscles 2 and 4 is normal in all cases.

Adults expressing Ten-mGD3620 under the control of Scer\GAL4elav.PLu (in the presence of Dcr-2Scer\UAS.cDa to increase the efficiency of RNAi) show significantly reduced avoidance of noxious temperature (46[o]C) compared to control flies.

Adults expressing Ten-mGD3620 under the control of Scer\GAL4elav.PLu (in the presence of Dcr-2Scer\UAS.cDa to increase the efficiency of RNAi) show no significant change in the kinetics of high temperature (46[o]C) induced paralysis compared to control flies.

Co-expression of both Ten-aGD8834 and Ten-mGD3620 under the control of either Scer\GAL4peb-GAL4 (expressed in the ORNs) or Scer\GAL4GH146 (expressed in the PNs) results in intermingling of Ncra\QFMz19.PH-expressing PN dendrites and Or47b-expressing ORN axons.

Co-expression of Ten-mGD3620 and Ten-aGD8834 under the control of Scer\GAL4Mef2.247 or Scer\GAL4nrv2.PS results in defects in T-bar ultrastructure, membrane organisation and apposition to contractile tissues.
