Expression of Lrrk3KD.Scer\UAS under the control of Scer\GAL4Mhc.PW does not result in any significant difference in amplitude of miniature excitatory junction currents (mEJCs), EJCs, or quantal content of larval neuromuscular junctions, as compared to controls.

The dendrites of dendritic arborizing neurons expressing Lrrk3KD.Scer\UAS under the control of Scer\GAL4ppk.PU exhibit a reduced proportion of stationary golgi outposts (GOPs) compared with controls. The dynamic GOPs that move in the anterograde direction (towards dendritic ends) show increase displacement and increased velocity. Retrograde displacement (towards cell bodies) and velocity are unaffected. The distribution of GOPs is shifted towards the distal region.

Expression of Lrrk3KD.Scer\UAS under the control of Scer\GAL4109(2)80 induces slight dendritic overgrowth compared with controls, mainly affecting higher order dendrites. Expressing Lrrk3KD.Scer\UAS specifically in Class IV da neurons under the control of Scer\GAL4ppk.PU also induces dendritic overgrowth.

Flies expressing Lrrk3KD.Scer\UAS under the control of Scer\GAL4ple.PG do not show a progressive reduction in electroretinogram amplitude (there is no significant difference in the amplitude seen in 3 and 28 day old flies).

Pre-synaptic or post-synaptic expression of Lrrk3KD.Scer\UAS (under the control of either Scer\GAL4elav-C155 or Scer\GAL4Mhc.PW) does not significantly affect the number of type I boutons compared to controls.

The number of dopaminergic neurons in the protocerebral posterior medial 1 and 2 clusters and in the protocerebral posterior lateral 1 cluster and the number of neurons in the protocerebral posterior medial 3 cluster is normal in 60 day old adults expressing LrrkY1383C.Scer\UAS under the control of Scer\GAL4ple.PF.

The dot-like small eye phenotype characteristic for flies expressing egrScer\UAS.cMa under the control of Scer\GAL4GMR.PFa is only slightly suppressed (or not at all) by co-expression of Lrrk3KD.Scer\UAS.

Co-expression of foxoScer\UAS.cPa and forP1.Scer\UAS under the control of Scer\GAL4GMR.PF results in degeneration of the eye. The severity of this phenotype is not suppressed by co-expression of Lrrk3KD.Scer\UAS.

Comments

Muscle or neuronal expression of Lrrk3KD.Scer\UAS (under the control of Scer\GAL4Mhc.PW or Scer\GAL4elav-C155 respectively) fails to rescue the synaptic morphology phenotype and average bouton size found in Lrrke03680 homozygous mutants.
