Expression of Gk1dsRNA.Scer\UAS driven by Scer\GAL4αTub84B.PL results in larval lethality ('Gk2dsRNA.Scer\UAS-let') or survival to adulthood ('Gk1dsRNA.Scer\UAS-sur'), depending on the insertion line. Expression of Gk1dsRNA.Scer\UAS-let (but not Gk1dsRNA.Scer\UAS-sur) lines driven by Scer\GAL4c564 or Scer\GAL4how-24B also results in larval and pupal lethality. Scer\GAL4c564>Gk1dsRNA.Scer\UAS-let third instar larvae often show melanotic masses before lethality mainly at the pharate adult stage (escapers have a curled or crumpled wing phenotype). Expression of Gk1dsRNA.Scer\UAS driven by Scer\GAL4GMR.PS or Scer\GAL4elav-C155 does not result in any obvious external or behavioral phenotypes.
Flies expressing GykdsRNA.Scer\UAS under the control of Scer\GAL4c564 have a lifespan similar to that of wild-type controls when raised on a sucrose only food source. However, when raised on a glycerol only food source, the mutant flies have a reduced lifespan compared to wild-type flies raised under these conditions, with the lifespan being similar to that under starvation conditions. The mutant flies show an intermediate lifespan when raised on a food source containing glycerol and sucrose.
Adult males expressing GykdsRNA.Scer\UAS under the control of Scer\GAL4c564 have a small but significant decrease in survivorship on a high salt diet (3.5% and 4.0%) compared to control flies.
