Duplication at the p53 locus. The 5' copy consists of wild-type exons 1 to 6, a partial insertion of pTV2 vector sequence, another copy of exon 6 and part of exon 7. The 3' copy lacks exon 1 and part of exon 2. A w+mW.hs marker and a single FRT site are located between the two copies.

γ-irradiated p53-ns/p53TM3 embryos and larval wing discs (the Tm3, Sb balancer has an inversion break point at the p53 gene), as well as γ-irradiated p53-ns homozygous wing discs, fail to show the expected increase in apoptosis.

Homozygous p53-ns flies show a reduction in survival following acute starvation during early adulthood (1-2 days). Reduced survival rates are seen in both males and females and also in 5-7 day old flies subjected to starvation. Similar effects on survival rates are seen in young adult flies or larvae exposed to complete nutrient deprivation, as well as larvae exposed to diluted food. Triacylglycerides (TAGs) and glycogen are used at higher rates in mutant flies exposed to nutrient deprivation compared to controls.

p53-ns mutant wing disc clones grow to a much larger size than control cell clones.

p53-ns mutant wing disc cells or fat body cells exhibit disrupted mitochondrial morphology, with a variety of shapes including globules and donut shapes as well as long filaments, as seen in wild-type.

p53-ns heterozygous mutants show apparently normal thorax structure.

Meiotic recombination frequency is reduced in p5311-1B-1/p53-ns and p532/p53-ns mutants compared with wild type.

p53-ns mutants are viable, but sickly with a shortened life span.

p53-ns mutants do not exhibit the normal increase in apoptosis following irradiation. However cell cycle arrest occurs as normal. Low doses of ionising radiation cause an increase in genomic instability and an increase in lethality when compared to controls.

Genetic Interactions
Statement
Reference

One copy of p53-ns suppresses the increase in survival rate in response to starvation seen in adult Dcr-1Q1147X/+ mutant flies.

A p53-ns heterozygous background strongly enhances the thorax disclosed phenotype induced by Nf-YAdsRNA.231-399.Scer\UAS.WIZ expression (under the control of Scer\GAL4pnr-MD237).

The rough eye phenotype caused by expression of DrefScer\UAS.cSa under the control of Scer\GAL4GMR.PS is suppressed by p53-ns/+ and by p53-ns/p53-ns (with stronger suppression in the homozygous background).

The mutant eye phenotype caused by expression of AIFΔN.Scer\UAS.T:Hsap\MYC under the control of Scer\GAL4ey.PH is not ameliorated if they are also homozygous for p53-ns.

Xenogenetic Interactions
Statement
Reference

The increase in the number of TUNEL-positive cells and the non-autonomous increase in BrdU incorporation seen in the wing discs in larvae expressing Rcom\RAcs2.Scer\UAS under the control of Scer\GAL4en-e16E at 30[o]C is largely suppressed by p53-ns.

The cell cycle arrest and undergrowth phenotype of 96 hour wing discs with posterior 'undead' Scer\GAL4hh-Gal4>hidScer\UAS.cYa, BacA\p35Scer\UAS.cHa cells is completely alleviated in both p535A-1-4 and p53-ns backgrounds.

Complementation and Rescue Data
Comments

Expression of p53GUS.PB under the control of Scer\GAL4yolk rescues the reduced survival seen in homozygous p53-ns flies in response to starvation.
